Objective To detect the C+33T polymorphism of IL4 gene promoter region and the total serum IgE levels of asthma children and normal children, and to explore the correlation between the C+33T polymorphism of IL-4 gene promoter region and the susceptibility of children with asthma and total serum IgE levels. Methods A C+33T polymorphism in the IL4 gene promoter region of 40 asthmatic children and 40 children without asthma was performed by polymerase chain reaction, restriction fragment length polymorphism(PCR-RFLP). The total serum IgE level of the children was detected by ELISA. Results ① The C allele frequency of +33 site of IL-4 gene promoter region in the asthma children were 22.5%, T allele frequency were 77.5%. The C allele frequency of +33 site of IL-4 gene promoter region in the normal children were 46.2% , T allele frequency were 53.8%. There was significantly different between two groups.( x2 = 9.5, x0.012 =9.2,respectively P<0.01);② Three genotype CC, CT, TT frequency of +33 site of IL4 gene promoter region in the asthma children were 7.5%, 25% , 67.5%. Three genotype CC、 CT、 TT frequency in the normal children were 35% , 22.5%, 42.5%.There was significantly different between two groups.(x2 were 10, x(0.01)(1) were 6.63,respectively P<0.01);③ The mean total serum IgE level of three genotype CC、 CT、 TT +33 site of IL-4 gene promoter in the asthma children of were 184±82 KU/L , 303.5±113 KU/L, 359±80 KU/L, respectively. The The mean total serum IgE level of three genotype CC, CT, TT of +33 site of IL-4 gene promoter in the normal children were 90±42 KU/L , 119±50KU/L, 145±38 KU/L respectively. There were significantly different between the genotype CC and CT or TT in the asthma children. (F=9.28>F0.01(2)(37)=7.35 P<0.01 .There was not different among the three genotype CC and CT and TT in the normal children;④ Mean total serum IgE level of different group of the same genotype CC and CT and TT in asthma children and normal children were significantly different (P<0.001)c Conclusions ①The C+33T polymorphism in the promoter region of IL4 gene is significantly associated with susceptibility of children asthma (P<0.01), the susceptibility of genotype CT and TT in the children asthma were elevated;(2) The C+33T polymorphism in the IL4 gene promoter region was associated with elevated total serum IgE levels ofasthma children, total serum IgE levels of genotype CT and TT in the asthma children were high. The T allele of +33 site of IL-4 gene promoter region in the asthma children were associated with high total serum IgE levels. T or C allele of+33 site of IL-4 gene promoter region in the normal children were not associated with high total serum IgE levels;(3) Mean total serum IgE level of different group of the same genotype CC and CT and TT in asthma children and normal were significantly different (P<0.001) .The results show children asthma gene genetic polyeffection.Postgraduate Liu ShiGuo (genetics) Directed by Vice-Pro.
