Astragalus and selenium on the Flammulina growth and applied research

In this study, Selenium and Astragalus were discussed on the growth and productivity of Flammulina, furthermore, the contents of Selenium in Selenium-added-Flammulina and Astragaloside in Flammulina raising in cultivation material adding Astragaloside were also surveyed and evaluated.With regard to the effect of Astragalus on Flammulina, at first, it was found that the culturing media with additional Astragalus could improve its yield to the peak by supplementation of 4% Astragalus decoction in the research of liquid fermentation. In the experiment of solid culture in large test tubes, the effect of Astragalus powder on the rate and tendency of growth were ascertained by measuring, and it was found that the rate was the fastest and the tendency of growth the best with 20% and 10% Astragalus addition. In the end, Astragalus divided in different proportions were added into the planting material to discussed its influence on industrialized cultivation of Flammulina. All the testes showed that supplementation of Astragalus into planting material coould shorten the growth cycle time and increase production.The days and the output of the blank control group(HO) were 79.17d and 35.30g respectively. But the days and the output of the treated H4 with 10% additional Astragalus were 70.13d and 48.83g respectively, compared with the blank control group,the growth cycle time shortened by 9.03 days and the yield of fruiting body improving by 38.33 %.Thin Layer Chromatography(TLC) was employed to tested whether Astragaloside was absorbed and transformed in Flammulina, and, the similar bands of standard showed that it worked.After that, determined of concentration of Astragalus in Flammulina by HPLC. Chromatographic conditions such as stationary phase(C18 column,250×4.6μm,5μm), mobile phase (100%acetonitrile), detection wavelength(203nm), column temperature(30℃) were identified. The determination results showed that: Astragaloside peak didn’t exist in HO, the contents of Astragalus in H5 is the most: 5.238mg/mL, and then H4 as well as H6,5.006 mg/mL and 4.897 mg/mL respectively.By means of various concentration supplementation of Selenium solution into Flammulina planting material, and then observation and recording of growth situation of Flammulina, it was found that Selenium can not only shorten its growth cycle time, but also increase the biomass, however, the height was only slightly affected.The contents of Selenium and ratio of organic Selenium in Selenium-added-Flammulina were determined by spectrophotometry. The ratio of organic Selenium in Flammulina was higher with the addition of Selenium-added-photosynthetic bacteria than that with the addition of inorganic Selenium mainly due to the organic transformation of Selenium by photosynthetic bacteria. At the same time, the conversion rate and food safety of Selenium were greatly enhanced.The Selenium absorbed by Flammulina with the supplementation of Selenium-added-photosynthetic bacteria was always higher than that with the supplementation of inorganic Selenium when the additional contents of Selenium was identical, however, the former is always lower than the latter in the contents of remaining Flammulina in the planting material. Ail of these proved that the Selenium transformed by photosynthetic bacteria was much more absorbable for Flammulina.
