Contents of this study are summarized as follows:1.Two TaqMan probe assays to detect Bifidobactirium and Bacillus were established, which both possessed favourable specificity and sensibility and were applied to detected Bifidobactirium and Bacillus quantitatively in digestive tract and respiratory tract of duck.2.FQ-PCR was applied to detect the amount of Bifidobacterium, bacillus, and Enterococcus in digestive tract and respiratory tract of duck in control group at 2 hour, 8 hour, 24 hour, 48 hour, 72 hour, 6 day, 12 day, 21 day, 36 day. Detection results showed that log10 16S rDNA copies of Bifidobacterium in whole esophagus and trachea were 3. 59 5. 95 and 3. 22 5. 70, log10 16S rDNA copies of Bifidobacterium (per gram of wet weight) in duodenum, jejunum, ileum, caecum, rectum were 5.94~7.64, 6.30~8.95,6.16~8.02, 7.51~9.40, 5.37~8.58; log10 16S rDNA copies of Bacillus in whole esophagus and trachea were 4.24~6.52 and 3.91~6.26, log10 16S rDNA copies of Bacillus (per gram of wet weight) in duodenum, jejunum, ileum, caecum, rectum were 6.58~8.01, 6.32~8.54,6.26~ 8.60,6.29~7.84,6.32~8.01; log10 23S rDNA copies of Enterococcus in whole esophagus and trachea were 3.52~6.08 and 4.42~7.06, log1023S rDNA copies of Enterococcus (per gram of wet weight) in duodenum, jejunum, ileum, caecum, rectum were 6.91~10.10, 6.53~8.71, 7.49~9.05, 7.78~9.84, 6.62~8.91。 3. FQ-PCR was applied to detect Bifidobacterium, bacillus, and Enterococcus quantitatively in digestive tract and respiratory tract of duck immunized with low duck plague virus by oral immunization, nose drop immunization, and subcutaneous immunization. Detection results showed that Bifidobacterium, Bacillus, and Enterococcus in digestive tract and respiratory tract of duck had various influence when immunized with low duck plague virus by various immunizations, the amount of three bacteriums were increased 10-fold than that of the control group in some day-age ducks.
