The Dynamic Changes of Serum Cytokines in Paraquat Poisoning Rats and the Artesunate Intervention Studies

Objective: In recent years,paraquat poisoning reportes are gradually increasing, which owns the feature of“toxic dose small, nor-effective antidote, nor-effective therapy currently”. After poisoning, the injury of lung is their outstanding performance, But the exact injury mechanism is not fully clear. In this study, by observing the content of cytokine interleukin -10 (IL-10), transforming growth factorβ1 (TGF-β1),which dynamic Changes in animal models with acute paraquat poisoning. To analyze the possible pulmonary damages mechanism induced by paraquat ,and evaluate their roles in the specific process. Moreover , we aimed to investigate effects of Chinese artesunate on pulmonary damages induced by paraquat and to observe the potential therapeutic effectes, so as to find and provide evidence for the treatment strategy of pulmonary damages induced by paraquat.Methods: A total of 70 SD male rats were randomly divided into two groups, Included a normal control group (20 rats) and an exposed group (50 rats).Then, exposed groups were divided into two groups: model group (25 rats,Referred to as the model group) and artesunate treatment group (25 rats,Referred to as the treatment group).After contamination, we will be divided into four observation periods, (that is observe at12,24,48 and 72 hours after exposure),Then ,we observe that the dynamic changes in serum IL-10 and TGF-β1 in different groups at different periods. We will analyze the results between different groups by analysis of variance, at the same time, we will slice pathological and observe pathological results in the different periods.Results: The SD rats showed poisoning symptoms starts at poisoning, the SD rats after exposure appears irritability, anorexia, malaise, lack of exercise, irritability, fluffy hair and shortness of breath etc,while the normal control group didn’t correspond a performance. the levels of Serum cytokines(IL-10 and TGF-β1) in the exposed rats had significantly increased start from 12 hours comparing to the normal control group, and peaked at 72 hours,their results were statistically significant in each time period(p<0.01). .Between the Artesunate treatment group and the model group, the levels of Serum in treatment group was less than the the model group start from 12 hours, but the statistically significant started after 48 hours (P <0.05). Compared the levels of Serum cytokines(IL-10 and TGF-β1) within each group of treatment group and model group at 12、24、48 and 72 hours,we find that the results were statistically significant in each group(P <0.05), but we must except the IL-10 in treatment group compared with 12 hours and 24 hours(P=0.053>0.05) . The pathological HE staining showed that the normal control group had no inflammatory cell infiltration, lung tissue integrity. The exposed group occurred alveolar congestion, edema, inflammatory cell infiltration since 12 hours after exposure, which reached to peak at 48 hours, then slightly reduced at the 72 hours, and the signs of lung tissue is degeneration of pulmonary fibrosis.The artesunate treatment group and the model group have similar performance, but the lung injury slightly reduced corresponds to the same time period since after 24 hours ..Conclusion: we fed paraquat to SD rats with a dose of paraquat 50mg/Kg, and diluted to 1ml,then we can copy toxic model. Paraquat poisoning may cause cytokine TGF-β1 and IL-10 uncontrolled release, The cytokine IL-10 and TGF-β1 may play an important roles in early stages of pulmonary damages induced by paraquat,Artesunate may influence the levels of cytokine TGF-β1 and IL-10 ,and then interfere with the course of acute paraquat poisoning, but the specific mechanism needs further study, and artesunate may play a protective effect, but the specific mechanism still needs further study.
