Para-nitrophenol is a kind of persistent organic pollutants,it had been used widespreadly in chemical engineering and pesticide industry in last century,and an intermediate in parathion-methyl catabolism.Its widespread distribution has raised people’s concerns on public health and environment protection.Bioaugmentation is a technology involved in addition of microbes,nutrition and metabolite analogue which has special function in facilitating sewage treatment.A strain nominated as LY2,which can degrade para-nitrophenol,was screened from a soil sample which was collected in Anyang insecticide factory,Henan.A 1,501bp fragment was obtained by PCR with a pair of universal primers of bacterial 16S rRNA,and has high homogeneity with Strains of Pseudomonas putida after blasted in NCBI.It is classified to P. putida in molecular Phylogeny,and nomenclatured as P. putida LY2.On the optimum conditions,when cultured in 30oC,150rpm,P. putida LY2 completely degrade 200mg/L PNP in 168h,but cannot was cultured in 300mg/L PNP.Induced in advanced with PNP in 4 different concentration can promote the degradation of PNP,both on time and concentration.P. putida LY2 can break down 500mg/L PNP in 72h drastically after treated with 200mg/L PNP 24hs.The addition ofβ-cyclodextrin also has synergic effect on degrading high concentration PNP. P. putida LY2 can break down 500mg/L PNP in 36h drastically when additive concentration withβ-cyclodextrin is up to 6mg/L.
