The Study of the Expression of LOX-1 in Aorta of Atherosclierotic Rabbits and the Intervention Effects of Ramipril

[Objective] To explore the expression of LOX-1 in aorta of atherosclerotic rabbits and the intervention effects of ramiril on the gene and protein expression of LOX-1 after establishing diertary atherosclerotic model, further elucidate the important role of LOX-1 during the development of atherosclerosis and provide a new thinking for the atherosclerotic treatment.[Methods] Thirty male New Zealand white rabbits were randomly and equally divided into three groups: control group(C group,n=10),fed with normal diet; atherosclerosis group(AS group,n=10), fed with high lipids diet and ramipril-treated group(RAMT group,n=10),fed with high lipids diet and treated with ramipril.They were fed for 16 weeks,the ascending aortae were harvested ,the concentrations of serum total cholesterol and low-density lipoprotein cholesterol were measured. We used hematoxylin and eosin (HE) staining to observe the morphology of ascending aortae and immunohistochemical staining to measure the expression of LOX-1 protein. The LOX-1 mRNA was analyzed by RT-PCR.[Results]1 、The morphology of the ascending aorta: the intima of ascending aorta in ramipril-treated group was thinner than that in AS group(P<0.05),and the lumina of ascending aorta in ramipril-treated group was larger than that in AS group(P<0.05),the intima structure of ascending aorta in C group was normal,there was no plaque in it.
