Anti-tumor Effects of Lupeol at Different Regimens and Its Role in Immune Systems

Objective: Lupeol,widely found in fruits and vegetables and other natural foods, has anti-tumor activity, as well as immune inhibitory function. To study the combinational effect of Lupeol on the tumor and immune system, we’ve set a long-term high-dose group, short-term high-dose group, the long-term low-dose group, and Negative control and positive control. The short-term high-dose group is set to imitate the process that Lupeol induced apoptosis of tumor cells, after stopping treatment, fragment of apoptotic cells activate the body’s immune systems, which could further eliminate tumor.Methods: Mice were inoculated subcutaneously with melanoma and were randomly divided into five groups: long-term high-dose group, short-term high-dose group, the long-term low-dose group, and the negative control and positive control. In addition to short-term group, mice were administrated with Lupeol for four consecutive days, other groups were administered three weeks, thrice a week. Record changes of mice’s body weight, activity, tumor growth during the administration. Spleen and blood was collect after the end of the experiment for flow analysis.Results: Compared with the control group, Lupeol exhibited almost no difference in body weight and daily behavior. According to the tumor growth curve, low dose in long term was more effective than other therapeutic regimen, followed by long-term high-dose group; high-dose short-term group did not show the desired effect. Mice treated with Lupeol short term at high dose generated an immune activationConclusion: Short-term high-dose group did exhibited an activation in immune system, but it showed a slight effect in tumor suppression, suggesting that in the anti-tumor process, Lupeol plays a dominant role, immune system may only play part of the supporting role. Lupeol was most effective at the long term small dose, providing experiment data for better regimen in further clinical applications.Objective: As Lupeol was extracted from natural food with very low toxicity, people can take it for everyday use. The anti-tumor effect of Lupeol has been confirmed, but there’s few research on its chemo-preventive effect. If we can confirmed that Lupeol have some preventive effect on tumor, it will be possible for us to reduce cancer through diet. The experiment mainly focused on the preemptive anti-tumor effect of LupeolMethods: 10 mice were randomly divided into two groups, control group and prevention group. After administrated orally for consecutive three weeks, we inoculated tumor cell s.c to the treated mice. During the following days we measured tumor size twice per week. At the same time body weight and daily activities of mice was also observed. Mice were sacrificed when the tumor growth to the ethical limits. Peripheral blood and spleen were collect for flow staining.Results: Three consecutive weeks’administration of Lupeol did not cause any change on body weight as well as daily activities of mice. Prevention group showed little inhibition in tumor growth, though the difference was not significant. Some immune cells of the prevention group was activated.Conclusion: Lupeol exhibited slight preventive effect and the effect is not obvious for its drug potency is relatively mild. As derived from natural food, Lupeol showed almost no toxicity. As a clinical preventive drug, Lupeol needs further development.Objective: To study the effect of Lupeol on the proliferation and apoptosis of myeloid leukemia cell lines K562, NB4, SHI-01, lymphoid leukemia cell lines Jurkat.Method: Cells were treated with Lupeol at five different concentrations in vitro. Proliferation inhibition was detected by MTT colorimetric assay, and cell apoptosis was Anti-tumor effects of Lupeol at different regimens and its role in immune systems Abstract analyzed by flow cytometry.Result: Lupeol significantly inhibited the proliferation of both myeloid and lymphoid leukemia cells, even reached 100% inhibition at the concentration of 300umol/L. Compared with negative control group, lupeol treated group showed a significant increase in cell apoptosis.Conclusion: Lupeol inhibited leukemia cell proliferation and induced cell apoptosis. This study provides evidence for the possible clinical application of lupeol in leukemia treatment.
