Objective:To investigave the DYS441, DYS442, DYS443, DYS444, DYS445, DYS446 STR loci allele distribution situation among male of Korean-Chinese nationality ethnic group and the Han nationality ethnic group in Yanbian area.Methods:(1)EDTA-theated blood samples were collected from 205 unrelated male of Korean-Chinese nationality and 207 unrelated male of the Han nationality,the DNA was extracted with blood genomic prep mini spin kit.(2)Six Y-STR loci were performed by using the techniques of PCR,polyacrylamide gel electrophoresis and silver staining.(3) The alleles of each Y-STR loci were sequenced, the allele ladders were prepared and nomenclature according to the recommendation of International Society of Forensic Genetics(ISFG).(4) The data were collected and analysed using statistics.Results:Total of 8,7,7,5,6,9 alleles were observed at DYS441, DYS442, DYS443, DYS444, DYS445, DYS446 loci in Chinese-Korean nationality, respectively. The calculated gene diversity(GD) were 0.7493,0.5083,0.6280,0.6969,0.5302,0.7897, respectively. A total of 152 haplotypes were found, and the haplotype diversity was 0.9501 in Chinese-Korean nationality. Total of 8,5,6,6,5,10 alleles were observed at DYS441, DYS442, DYS443, DYS444, DYS445, DYS446 loci in Han nationality, respectively. The calculated gene diversity (GD) were 0.7493,0.5083,0.6280,0.6969, 0.5302,0.7897, respectively. A total of 174 haplotypes were found, and the haplotype diversity was 0.9880, in Han nationality.Conclusion:DYS441, DYS442, DYS443, DYS444, DYS445 and DYS446 loci has highly genetic polymorphism and stability in Chinese-Korean and Chinese-Han. The data obtained can be used in medical jurisprudence individual identification, paternity test and population genetics investigation for Chinese-Korean nationality ethnic group and Han ethnic group.
