Objective: To study the PD98059 on proliferation of human gastric cancer cell SGC-7901 cell lines apoptosis and expression of MMP2 / 9 , and further explore its possible mechanism . Methods : application of cell culture techniques in cultured human gastric cancer cell SGC - 7901 , detected by MTT PD98059 inhibition of cell killing ; immunohistochemical assay of gastric cancer cells p-ERK protein expression ; the flow cytometry gastric cells wither death changes ; RT - PCR assay in gastric cancer cell MMP - 2 , MMP -9 expression . Results : PD98059 can inhibit the growth and proliferation of SGC-7901 cells in a dose - time dual- effect relationship ; Immunohistochemistry showed decreased expression of p-ERK PD98059 treated SGC-7901 cells ; flow cytometry show SGC-7901 cells apoptosis and increased with increasing drug concentration (Annixin V method) ; MMP9 expression in gastric cancer cells was significantly decreased after PD98059 treatment of MMP2 expression decline is not obvious . Conclusion : PD98059 can inhibit the growth of human gastric cancer SGC-7901 cells proliferation and induce apoptosis ; MAPK -ERK signaling pathway involved in the expression of MMP2 / 9 in the regulation of SGC-7901 cells by inhibiting the activation of the signaling pathway , significantly inhibited MMP9 expression of MMP2 expression inhibits obvious .
