Research on the Materials of Anti-virus of Gegenqinlian Decoction

Master candidate: Gong Zhanwen Supervisors: Professor Shi Renbing(School of Chinese pharmacology,Beijing University of Traditional Chinese Medicine,Beijing 100102)This dissertation includes three parts: summary of related literatures,studies on the materials of Gegenqinlian decoction effective against virus and discussion of the experiments.Part one:Literatures and summarizations about Gegenqinlian Decoction.Documents about the research progresses in the chemical constituents and pharmacological activities of Gegenqinlian Decoction and its single medicines in last five years were summed up and analyzed. 205 references were cited.Part two:Exprriments studies about Gegenqinlian Decoction.The effective fractions of Gegenqinlian Decoction are extracted and separated .Nine compounds are isolated with silica gel, polyamide, sephadex gel chromatography and six compounds are identified with 1HNMR and 13CNMR spectra. They are four flavonoids (purarin, baicalin, wogonin, isoliquiritigenin) and two alkanoids(berberine,jatrorrhizine).All of six compounds are firstly obtained from Gegenqinlian Decoction.HPLC figs of the effective fractions of Gegenqinlian Decoction are established primarily. By analyzing and comparing,34 chromatographic peaks are found in the HPLC figs of the effective fractions of Gegenqinlian<WP=7>Decoction and the ascriptions of the peaks are validated in the HPLC figs of every single medicine. Fourteen Peaks are confirmed after comparing with standard samples.Major compositions of the effective fractions of Gegenqinlian Decoction Were defined as alkaloids and flavonoids, their content were determined to be 12.61% and 28.77% respectively. The linear ranges of berberine and baicalin were 0.0025-0.0125mg/mL and 0.01416-0.04248mg/mL respectively, both of them gave the corralation coefficient 0.9997 and 0.9995.The average recoverie of baicalin was 102.0%(RSD2.75%,n=5).The contents of index compounds including baicalin and puerarin ere determined by HPLC, their content were determined to be 8.70% and 7.04% respectively. The calibration curve were linear at a range of 0.26-2.08μg (for baicalin, r=0.9997) and 0.5-4.0μg (for puerarin, r=0.9998) respectively. The average recoveries were 98.61% (RSD2.77%, n=5) and 102.4% (RSD2.70%, n=5) respectively.Part three: analysis and discussion of experiments.
