Curcumin Inhibits the Proliferation of Airway Smooth Muscle Cells in Airway Remodeling and Its Mechanism

Objective: airway smooth muscle layer thickening of the pathological changes is important pathological features of airway remodeling. The study found that airway remodeling in asthma patients (especially with moderate to severe asthma), an increase in the number of airway smooth muscle cells (hyperplasia) or cell hypertrophy, which leads to the thickening of the smooth muscle layer. Factors thickening of airway smooth muscle cells (Airway smooth, Muscle cells ASMCs was examined) hyperplasia is predominant. This study was prepared BALB / c mice with chronic asthma airway remodeling model to study the inhibitory effect of curcumin on proliferation of ASMCs, to explore the possible signal transduction mechanisms and drug targets. : Ovalbumin (OVA) sensitization and challenge of SPF BALB / c mice to establish mice with chronic asthma airway remodeling model. The mice were observed when excited symptoms; detection of airway responsiveness; ELISA assay of serum specific IgE content; eosinophils accounted for a percentage of the total number of white blood cell counts in bronchoalveolar lavage fluid (BALF); pathological observation airway structural changes ; using immunohistochemical identification of airway smooth muscle and image analysis software IPP measured bronchial wall thickness and the thickness of the smooth muscle layer. Results: 1 mouse model of excitation early irritability, shortness of breath, salivation, abdominal cramps and incontinence positive, curcumin prevention and treatment groups obvious symptoms, symptoms late, and to ease soon, asymptomatic normal control group. 2 curcumin prevention and treatment groups airway reactivity was significantly lower than the asthma group, P lt; 0.05, but still higher than the control group, P lt; 0.05. 3. Of curcumin Prevention and treatment groups serum specific IgE was significantly lower than the asthma group, P lt; 0.05, but still higher than the control group, P lt; 0.05 4. curcumin prevention and treatment groups in BALF eosinophils accounted for a percentage of the total number of white blood cells were significantly lower than asthma group, P lt; 0.05, but still higher than the control group, P lt; 0.05. 5 bronchial wall thickness of curcumin prevention and treatment groups, the bronchial smooth muscle layer thickness and number of bronchial smooth muscle cells were significantly lower than the asthma group, P lt; 0.05, but still higher than that in control group, P lt; 0.05. Conclusion: 1. Successfully established airway remodeling model. 2 curcumin can significantly reduce activity in mice, airway hyperresponsiveness, serum-specific IgE in BALF fluid eosinophils accounted for a percentage of the total number of leukocytes and reversing airway remodeling pathological changes. Curcumin can inhibit airway remodeling model of airway smooth muscle proliferation. Curcumin inhibition of proliferation of ASMCs may be associated with the inhibition of ERK1 / 2 signaling pathway.
