Biochemical Pharmacological Study of Polysaccharide from Pilose Antler Against Liver Injuries

Pilose antler is unossified deer horn of the male spotted deer(Cervus nippon Temminck var. mantchuricus Swinhoe) or red deer(Cervus elaphus L.), and an famous and valuable Chinese traditional medicine. It takes on abroad pharmacyological activities, for instance, regulating effects on cardiacvascular system, tonocity function, hormone-like action, promoting effect on healing of wound, anti-aging action. However, the effect and mechanism of pilose antler aginst liver injuries has not been clarified. We have studied the effect and mechanism of the polysaccharide from pilose antler(PSPA) against liver injuries, in order to elucidate new pharmacological action and new medicinal value, and furnish the theoretical basis for developing and exploiting new drug from deer antler against liver injuries .To investigate the effect of PSPA against experimental liver injury, models of liver injury in mice were selected, such as the model mice of liver injury induced by carbon tetrachloride (CCl4), thioacetamide(TAA)acetaminophen(AAP),D-galactosamine(D-Gla)(similar with virus hepatitis), ethanol, ethionine(fat liver), and naphthyl isothiocyanate(ANIT) (acute jaundice hepatictis).The <WP=47>effect of PSPA on experimental liver injury was observed. It was demonstrated that PSPA lower the serum SGPT activity significantly in the model mice of liver injury induced by CCl4, AAP and TAA, increase the contents of liver DNA, RNA, protein, glycogen significantly, and decrease the liver TG level significantly in the model mice of liver injury induced by CCl4. It was indicated that PSPA exhibits the action against experimental live injury PSPA lowered the serum SGPT activity significantly in the model mice of liver injury induced by D-Gla, It was indicated that PSPA exhibits the action against live injury similar with virus hepatitis.PSPA lowered the serum SGPT activity and decrease the liver TG level significantly in the model mice of liver injury induced by ethanol and ethionine, It was indicated that PSPA exhibits the action against live injury induced by ethanol and similar with fat liver.PSPA could lower the serum SGPT activity and total bilirubin (TBIL) level, increase the content of DNA, RNA, protein, glycogen in liver, decrease the liver triglycerides(TG) level significantly, and enhence the tendency of bile excretion in the model mice of liver injury induced by ANIT. It was indicated the PSPA exhibits the action against live injury of acute jaundice hepatictis induced by <WP=48>ANIT,and furnish the pharmacological basis of therapy PSPA for acute jaundice hepatictis .To investigate the mechanism of action of PSPA against liver injuries, the anti-lipoperoxidation of PSPA in mice liver was studied.The model mice of lipid peroxidation induced by CCl4, ethanol, AAP was selected.It was demonstrated that PSPA decrease the content of MDA from lipid peroxidation in mice liver induced by CCl4, ethanol, AAP.In additon,PSPA could enhance the activity of antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase(GSHpx) in mice,it can help to eliminate free radical generated from body.Furthermore,PSPA could increase the content of liver microsomal cytochrome P-450 in mice,it was indicated that PSPA induce the“enzyme”in liver microsomal,and enhance the detoxification of liver.The above mentioned results indicated that PSPA show obvious effect against liver injury, the mechanism of action was related to the fellowing:(1)Anti-lipoperoxidation;(2)Maintaining the metabolize normalization of nucleic acid,protein ang sugar;(3)Inducing liver microsomal the“enzyme”(cytochrome P-450), enhancing the detoxification of liver;(4)Enhancing the activity of <WP=49>antioxidant enzymes, such as SOD, CAT and GSHpx.We make further study on the mechanism of action of PSPA against liver injury. The effects of PSPA on the lipid peroxidation [ malondialdehyde (MDA) formation] in microsomes of rat brain, liver and kidney induced by NADPH- Vitamine C and ferrous- cysteine systems in vitro
