The Effect of Gingivitis on Cyclosporine A-induced Gingival Overgrowth

Objective: The study of cyclosporine A (CsA) and endotoxin (LPS) in cultured human gingival fibroblasts (GFs) the impact of research through the establishment of a rat model of CsA-induced gingival hyperplasia gingival inflammation gingival hyperplasia . Cell no significant change after CsA or LPS role morphology, CsA can promote cell proliferation and secretion of TGF-β1 of LPS effect and its opposite, the combination effects can significantly promote cell growth and secretion of TGF-β1, than the simple use of CsA The effect is more obvious. 3 tooth neck ligation wire can successfully established periodontitis model in rats, after one week ligation can occur gums gingivitis, in the performance of the periodontitis in the first 4 weeks. 4. Use of CsA oil solution gavage nine weeks, and the successful establishment of rat model of drug-induced gingival hyperplasia, weight loss medication rats before and after teeth the gums are obviously hyperplasia, gingival hyperplasia is more pronounced inflammatory drug group. After treatment in rats than untreated rat serum significantly increased expression of TGF-β1, and then tie on serum TGF-β1 and has no effect. Conclusion: 1.CsA GFs proliferation and secretion of TGF-β1 and LPS have synergistic effect. 2. Tooth neck ligation wire method can be successfully established periodontitis models can occur after 1 week gingivitis. Inflammatory presence of CsA-induced gingival hyperplasia will be more obvious, TGF-β1 may be important factor in the pathogenesis.
