Research on the Effect and Mechanism of Chitosan to Dry Eyes

Objective To explore the chitosan mechanism of action of the treatment of dry eye . 32 New Zealand white rabbits (weighing 2 ± 0.5 Kg) were randomly divided into normal control group , dry eye group , chitin Sugar group and tears ran group . Production of animal models of dry eye , the chitosan group and the tears ran group topical 2% PBS solution with chitosan solution and tears ran topical eye drops , dry eye group and normal control group . 1, 2, 3, and 4 weeks after the medication , and observe the change of appearance of each group hotels Tujiao conjunctival , simultaneous detection of the Schirmer test value , but values ??. Each group were four eyes, the animals were sacrificed at each time point , take the conjunctival tissue slices Immunohistochemistry was used to detect the conjunctiva the Muc-5ac protein expression changes . And the first 4 weeks after the administration , each group were conjunctival tissue homogenates extracted protein solution , semi-quantitative detection with the Western Blot method wherein protein Muc-5ac . Chitosan group and the tears ran group after three weeks of topical Schirmer test values ??and BUT values ??than those of dry eye group increased significantly (P lt; 0.01). Chitosan group and tears contingent group after treatment compared with dry eye syndrome group conjunctival epithelial mucin Muc - 5ac grayscale value , protein bar band optical density (OD) increased ( P lt; 0.05 ) , and similar to the normal control group ( P gt; 0.05). Chitosan group with tears contingent group Schirmer test , BUT value , conjunctival epithelial mucin Muc - 5ac gray value , protein bands of the optical density (OD) of all indices showed no significant difference (P gt; 0.05). Conclusion Chitosan can promote conjunctival epithelial ( goblet cells ) to produce mucin MUC-5AC treatment of dry eye .
