Radiation Protective Effects of D.candidum on GM0639 Cells

Objective: In order to evaluate D.candidum radiation protective by cell experiments.Methods :MTT method was used to check the effect of different concentration and treatment time of D.candidum to cytotoxicity. Cytokinesis block micronucleus method (CB method) was used to determine the micronucleus frequency (MNF) and the micronucleus cell frequency (MNCF) of cells treated by D.candidum additive and with different doses of 60Coγ-rays. Flow cytometry(FCM) and propidium iodide(PI) staining was used to measure the effects of D.candidum on cell cycle phases which exposed to 4Gy 60Coγirradiation.The apoptosis rates of GM0639 cells were determined by AnnexinV-FITC/PI double staining .Results:The experiment results show D.candidum has no cytotoxicity in the range of 0~100mg/mL to GM0639 cells .When drug concentration is 60mg/mL and treatment time is 24 h, the dose-response curves of MNF and MNCF of irradiated cells combined with treatment of or not D.candidum additive confirm that MNF and MNCF of cells treated by D.candidum are both significantly lower than that treated by radiation alone.The results of PI staining indicate that the proportion of SubG1 of GM0639 cells after 4Gy 60Coγirradiation were markedly higher than that of control group (p<0.05).The proportion of SubG1 was decreased from 33.14±2.11% in the irradiation treated cells to 5.57±1.02% in the 100mg/mL of D.candidum and irradiation treated cells in 24h(p<0.05).The results of apoptosis of GM0639 cells showed that D.candidum can inhabit the apoptosis which induce by irradiation,after treated with 100mg/mL of D.candidum and irradiation ,the apoptosis rates decreased from 20.94±0.32% to 4.54±0.32%(p<0.05).Conclusion:The cell experiments demonstrated that D. Officinale has radiation protective effects on GM0639 cells .
