Early Evaluation Treatment Efficacy of Thermochemotherapy on Colon Cancer Cells HCT116 by 18F-FDG Uptake Rate

Inhibitory effect of 5-FU in the uptake of 18F-FDG on human colon carcinoma Cells 116Purpose:To establish a stable method to detect the 18F-Fluoro-deoxyglucose (18F-FDG) uptake in HCT-116 cell, and using 18F-FDG uptake inhibition rate to evaluation early response of heat therapy, chemotherapy, and thermo-chemotherapy on colon cancer cell HCT-116.Methods: Uptake rate of 18F-FDG in HCT116 were determined when the experimental conditions were changed in turn, including cell concentration, reaction time, 18F-FDG radioactive activity, and glucose concentrations incubation in 37℃. Before and after giving different concentrations 5-FU, the MTT method was adopted to measure cell proliferation inhibition rate and 18F-FDG uptake rates were also measured.Results: Uptake rate of 18F-FDG was （43.77±4.16）% at the condition of 1×106 cells, 18F-FDG radioactive activity of 3.7 KBq, glucose concentration of 5.5mmol/L and 100min incubation at 37℃. 24h after administration of 5-FU, the Uptake rates of 18F-FDG were negatively correlated with the dose of 5-FU （r=-0.879,P<0.01）, and the 18F-FDG uptake inhibition rates were positively correlated with the cell proliferation inhibition rates （r = 0.831, P < 0.01）.The binding efficiency of 18F-FDG was (43L glucose concentration,and 100min incubation at 37℃. The binding efficiency and uptake inhibition rates of 18F-FDG were was（32.47±5.43）%,（26.44±3.62）%,（22.26±3.63）%,（16.10±6.40）% and（25.81±5.64）%、（39.58±3.74）%、（49.31±5.16）.77±4.16)% at the condition of 1×106cells, 3.7 KBq 18F-FDG, 5.5mmol/%、（ 62.27±7.50 ） % at 24h after the administration of 10, 20 ,30 and 50μg/ml 5-FU , respectively. The 18F-FDG uptake was negatively correlated with the dose of 5-FU（r=-0.879,P<0.01）. The uptake inhibition rate was positively correlated with the proliferation inhibition （r = 0.831, P < 0.01）.Conclusion: The binding efficiency of 18F-FDG on HCT116 decreased at 24h after administration of 5-FU .18F - FDG uptake is expected to reflect the inhibitory effect of 5-FU on colon cancer as early as 24h.Early evaluation treatment efficacy of thermochemotherapy on colon cancer cells HCT - 116 by 18F-FDG uptake ratePurpose: The first purpose of this study is to investigate the influence of heating therapy to the sensitivity of tumor cell on chemotherapy drug and the other is whether the 18F-FDG as the common imaging agent can evaluate thermo-chemotherapy effectiveness in early phase post-procedure.Methods: Before and after giving chemotherapy （100μg/ml 5-FU, 10μl）, thermotherapy （43℃, 30min） and thermo- chemotherapy （combining two conditions, first thermotherapy, then 5-FU） at 24h or 48h, cell proliferation inhibition rate and 18F-FDG uptake inhibition rates were measured. The degree of cellular apoptosis was determined by flow cytometry, and the cell ultra structure was observed by transmission electron microscope. The glucose concentration and lactate dehydrogenase concentration in cell culture of supernatant fluid before and after treatment were measured.Results: After treated with thermotherapy, chemotherapy, and thermo-chemotherapy at 24h and 48h,cell proliferation inhibition rates were （10.24±8.00）%, （25.68±6.10）%, （20.67±7.24）% and（19.38±5.49）%（39.24±7.83）% （42.71±9.59）% respectively, and the 18F-FDG uptake inhibition rates were （12.94±2.80）%, （28.25±4.59）%, （21.60±3.68）% and（25.47±7.09）%（37.18±5.16）% （39.33±9.19）%. early apoptosis rate by flow cytometric analysis testing at 24 hours was（9.80±0.16）%,（19.80±2.40）%,（15.70±1.80）%. After treatment early apoptosis showed by transmission electron microscope, and glucose concentration and lactate dehydrogenase concentration in the cell culture medium were higher than before treatment. The inhibition rate of cellular proliferation was measured by the method of the binding efficency of 18F-FDG uptake ,which was correlated with flow cytometry and MTT assay , and by test before and after treatment the two time periods with statistical significance （P <0.05）, but there was not statistically significant of the comparison between thermal chemotherapy and chemotherapy group（P> 0.05）.Conclusion: 18F-FDG uptake inhibition rate can evaluated early effect of thermotherapy and chemotherapy, and there was no synergistic effect by combining 43℃heat treatment 40min to low concentration 5-FU on human colon cancer cell HCT - 116.
