Method of all specimens from archived paraffin blocks of surgical specimens of breast cancer in the Department of Surgical Oncology in China from January 2001 to August 2007, a total of 60 cases, including 30 cases of familial breast cancer, 30 cases of sporadic breast cancer. Familial breast cancer group entry criteria to the first-degree relatives of breast cancer patients have a family history of breast cancer, sporadic breast cancer in breast cancer patients with no family history of breast cancer as the inclusion criteria. All patients were female patients, pathological invasive ductal carcinoma, and put without preoperative chemotherapy, age range 41 to 65 years old. BRCA1 monoclonal antibody, mouse anti-human SP kit, DAB were purchased from Fuzhou, the development of new biotechnology companies. FANCD2 antibody was purchased from SANTA CRUZ biotechnology development company. To complete the whole process of the experiment in Gastric Cancer Research Center of China Medical University, The First Affiliated Hospital. Detected by immunohistochemistry SP method. All wax block after slicing, after 60 ℃ drying, dewaxing and hydration, hydrogen peroxide closed endogenous peroxidase, high temperature and high pressure treatment, antigen retrieval, cooling, adding peroxidase blocking agent A, serum closed, join an anti-B and anti-C 10 ~~ 15min of D reagents, and finally DAB stain, hematoxylin, dehydrated cementing light microscope. Observed BRCA1 and FANCD2 expression in familial and sporadic breast tissue to discuss familial and sporadic breast cancer in BRCA1 and FANCD2 expression differences and their correlation. Statistical analysis using SPSS software to complete. 1, BRCA1 protein expression in familial breast cancer in 21 cases (70.0%, 21/30), and expression in sporadic breast cancer cases (13.3%, 4/30), between the two groups was statistically differences (P <0.05). 2, FANCD2 protein expression in 12 cases (40.0%, 12/30), the expression in sporadic breast cancer, 20 cases (66.7%, 20/30) between the two groups was statistically difference in familial breast cancer (P <0.05). 3 familial breast cancer in BRCA1 mutation group, FANCD2 expression in 11 cases (36.7%, 11/30), expressed in BRCA1 mutation is not the group 1 patients (3.3%, 1/30), between the two groups is statistically there are differences (P <0.05). 4 sporadic breast cancer, FANCD2 expression in BRCA1 mutation group 4 cases (13.3%, 4/30), the expression in the BRCA1 mutation is not the group of 16 patients (53.3%, 16/30) between the two groups is statistically was no significant difference (P> 0.05). Conclusion 1, BRCA1 expression in familial breast cancer is higher than the expression in sporadic breast cancer, suggesting that BRCA1 plays a very important role in familial breast cancer. 2, FANCD2 gene expression differences exist in familial breast cancer with sporadic breast cancer, high expression in sporadic breast. Tip FANCD2 gene as a tumor suppressor gene in breast cancer play a role in the formation process. Of FANCD2 gene BRCA1 expression correlation in familial breast cancer, a negative correlation. Mentioning demonstration Fanconi anemia / BRCA pathway plays a certain role in the formation of breast cancer especially in familial breast cancer. 4, according to the the FANCD2 gene BRCA1 gene expression can understand the familial susceptibility to breast cancer risk and normal people sick.
