This test were used organic solvents oscillation extraction technology, ultrasonic enhanced extraction technique, microwave-assisted extraction technology three methods of resveratrol in grape pomace, reuse macroporous resin adsorption chromatography resveratrol crude purified, refined, and the extraction and purification of process parameters for the selection and optimization to obtain a high purity product obtained the optimum operating parameters of each method, and using liquid chromatography Resveratrol content of the sample raised and analyze the chemical composition, the intention to provide a theoretical reference for the development and utilization of resveratrol in grape pomace. Solvent oscillation resveratrol with ethanol as extracting agent. On the basis of single factor test, L 9 (3 4 ) orthogonal experiment shows that the ethanol concentration of the solution the oscillation extract resveratrol extraction effect greater impact , reached a very significant level, and primary and secondary order of ethanol concentration> extraction temperature> solid to liquid ratio> extraction time. The results showed that the extraction temperature of 70 ° C, the solid-liquid ratio of 1:35 g / ml, the ethanol concentration of 60%, pH 4, the extraction time of 4 h. Under optimum conditions, the extraction rate of up to 0.165%. Ultrasonic extraction of resveratrol with ethanol as the extraction solvent. On the basis of a single factor, the L 16 (4 5 ) orthogonal experiment to determine the optimum process parameters as follows: 60% ethanol concentration, solid-liquid ratio of 1: 25 g / ml, the duty cycle of 1:2, ultrasonic time of 6min ultrasound 500W power, in this condition, the extraction rate of 0.277%. Its range analysis and variance analysis, results show that the orthogonal experiment examined five factors of ultrasound to strengthen the extracted resveratrol primary and secondary order ultrasound time> duty cycle> ultrasonic power solid-liquid ratio> ethanol concentration, and reached a very significant level of ultrasonic time on the extraction effect, duty cycle, and ultrasonic power on the extraction effect also reached significant levels. Microwave assisted extraction of resveratrol with ethanol as the extraction solvent. On the basis of single factor test, L 9 (3 4 ) orthogonal experiment results showed that: the orthogonal experiment examined three factors microwave assisted extraction white veratryl alcohol, primary and secondary order microwave time> microwave power> solid to liquid ratio, and the microwave time reached a very significant level of extraction effect. The best process parameters for microwave assisted extraction of resveratrol: the microwave time as 70s, solid-liquid ratio of 1:20 g / ml, microwave power 600W Under these conditions, the extraction rate of up to 0.284%. Compare the results of three methods: consider the composition and content of the extract, the solvent oscillation ingredients extracted complex purification have been affected on the back of the extracts, and low efficiency, time-consuming. The other two methods, the extract component is basically the same, and high efficiency. The three methods are almost no solvent residue chemical separation methods, reliable operation process, considering the ultrasonic extraction is more appropriate for large-scale production. Macroporous resin adsorption chromatography purification of resveratrol, considering the rate of adsorption and desorption rate of resveratrol extract AB-8 is more appropriate value. Selected on the sample liquid flow rate of 1.5 BV / h, the sample concentration of 10mg/ml, on the sample solution to pH 4, and the desorption solution concentration of 80% ethanol and desorption the flow rate 0.75BV / h, as well as the regeneration of the resin, and many other factors directly affect the chromatography results. Resveratrol sample will be extracted and purified by high-performance liquid chromatography detection, seen through the HPLC chromatogram of the samples and standards, the liquid chromatogram and standard chromatogram peaks situation roughly the same. After purification, resveratrol was 0.81%, a purity of 42.5%. Otherwise other absorption peaks, may be affected by the interference of a small amount of impurities.
