The Content Change of EGF and TGFα, and the Expression of EGFR by Shuganhewei Pill in Stress-related Rats

Objective: To study the protective effect of the method of the clinical empirical prescription Shuganhewei Pill on stressed-peptic ulcer, acute or chronic gastritis and its mechanisms.Methods: The rats were randomly divided into four groups: normal control group(group A), model group(group B), Shuganhewei Pill group(group C), ranitidine group(groupD). Group C and groupD were instilled with Shuganhewei Pill ( 1.08g/kg·d ) and ranitidine (0.0015g/kg·d) respectively for 7 days, group A and group B were instilled with the same volume physiological saline for 7 days. After 7 days, rats in group B、C、D were stress- induced gastric mucosal lesion rat model. Then, each group measured these indexes separately:①gastric mucosal ulcer index(UI);②gastric mucosal blood flow(GMBF);③changes of microstructure histopath- ologic gastric mucosal tissue;④content of PGE2PGI2、EGF and TGFa by Radioim- munoassay;⑤proteinic expression of EGFR by immunohistoch- emistry;⑥Gene expression of EGFR by In situ hybridization(ISH) Results:1、Compared with model group, Shuganhewei Pill group、ranitidine group can reduce UI obviously(P<0.01); compared with ranitidine group, Shuganhewei Pill group can reduce UI obviously(P<0.05).2、Compared with model group, Shuganhewei Pill a groups ranitidine group can increase GMBF obviously(P<0.01); compared with ranitidine 1 group, Shuganhewei Pill group can increase GMBF obviously(P<0.05).3、Compared with model group, Shuganhewei Pill group、ranitidine group can obviously improve gastric ulcer rats’ gastric mucous membrane micro-structure.4、Compared with model group, Shuganhewei Pill group、ranitidine group can obviously improve PGE2 in gastric mucous membrane tissue and PGI2 in blood serum.5、Compared with model group, Shuganhewei Pill group、ranitidine group can obviously improve EGF、TGFαin blood serum and gastric mucous membrane tissue and have statistics differences(P<0.01); compared with ranitidine group, Shuganhewei Pill group can obviously improve EGF、TGFαin blood serum and gastric mucous membrane tissue and have statistics differences(P<0.05).6、Compared with model group, Shuganhewei Pill group、ranitidine group can increase the proteinic and gene expression of the co-receptor of EGF and TGFαin the gastric mucosal tissue , it has extremely prominent difference (P<0.01); compared with ranitidine group, Shuganhewei Pillgroup can increase the proteinic and gene expression of the co-receptor ofEGF and TGFαin the gastric mucosal tissue , (P<0.05).Conclusion:1、Shuganhewei Pill can reduce rats’UI with irritable gastric mucousmembrane damages, it has better protective action on the animal’s gastricmucous membrane.2、Shuganhewei Pill can improve the GMBF in gastric mucous membrane,raise the PG、EGF and relevant peptide material in vivo, and content ofEGFR.3、The above-mentioned functions show the protective effect ofShuganhewei Pill may be related to improving the synthesize and releaseof the endogenetic and protective PG, and improving the synthesize of thegrowth factor with improving the growth of the membrane of stomachand the expression of their receptor, and to make the effect ofconfrontating the injury of the membrane of stomach.
