The Effect of Pamd to P53 and Caspase-3 mRNA Expression on Tumor Tissue of S180 Bearing Mice

Objective:To review the effect of the anti-tumor and immune function on tumor bearing mice,through research antineoplasmic activity of PAMD in internal experiment; and through test mRNA expression of P53 and Caspase-3 in tissue of tumor,to approach the possible mechanism of action about the anti-tumor growth and development.Methods:The metastatic tumor model of tumor-bearing C57BL/6 mice were establish-ed with S180 cell line by hypodermic of right oxter. The model mice were divided into 5 groups at random, PAMDhigh-do se,mid-dose, low-dose group, positive control group and model group.To observe the effect on the basal survival condition of tumor bearing mice and calculate inhibition rate of tumor,index of the spleen and index of the thymus gland according to formula.The reverse transcripttion polymerase chain reaction (RT-PCR)were applied to detect the differences of P53 and Caspase-3 gene expression in the tissue of tumor, to approach the mechanism of action about the anti-tumor growth and development.ResultsrThe anti-tumor rate of PAMD high, mid,low dose groups were 31.47%,44.11%,38.37%,all exceedded effective anti-tumor rate 30%. Compared with the model group, the spleen index and the thymus gland index of PAMD groups didn’t have statistical significance(P>0.5). PAMD could upwards-regulate the expression of gene of P53 and Caspase-3 mRNA in tumor tissues, compared with the model group with statistical significance(.P<0.05).Conclusions:PAMD could inhibit evidently the tumor growth in tumor-bearing mice of S180 cell line.The effect of inhibition was possib ly related by regulating immunologic function, upwards-regulate the expr ession of gene P53 and Caspase-3mRNA, to induce programmed cell death so that it could inhibit the growth of tumor.
