Matrix Metalloproteinase-3 and Macrophage Colony-Stimulating Factor Correlate with Disease Activity and Mechanism in Ankylosing Spondylitis

Objective: The gene chip technology found matrix metalloproteinase -3 (MMP-3) and macrophage colony-stimulating factor (M-CSF) increased expression in the synovial tissue of patients with spondyloarthropathy (Spondyloarthropathy, SpA) , through the detection ankylosing spine spondylitis (Ankylosing Spondylitis, AS) in serum and synovial fluid MMP -3 and M-CSF expression levels and correlation analysis with AS disease activity in patients to discuss their role in the pathogenesis of AS . Study the possible interaction between MMP-3 and tumor necrosis factor -α (TNF-α) , and explore their role in the pathogenesis of AS . Methods : Peripheral blood mononuclear cells (PBMC) from 11 patients with SpA synovial tissue of patients and 10 normal control individuals extracted total RNA, using the the 1176 gene lattice oligonucleotide microarray technology to detect the gene expression spectrum. Simultaneously the two groups of patients with AS serum MMP -3 and M-CSF expression levels were measured by ELISA , a group of 41 patients without any treatment of anti - TNF-α biologics AS serum , another group of 13 patients with the application anti - TNF-α monoclonal antibody Infliximab treatment and serum of patients with AS after 14 weeks of treatment , while the contrast detection and 28 normal controls individual serum and 15 patients with SpA synovial fluid of patients with MMP-3 and M -CSF levels . And analysis of 41 patients with AS serum MMP-3 and M-CSF expression levels with disease activity indicators such as ankylosing spondylitis disease activity index (BASDAI) Ankylosing Spondylitis Functional Index (BASFI ) , ESR and CRP between correlation. Application having the cleavage of the active region of human recombinant MMP-3 (Human recombinant MMP-3, MMP-3) in vitro stimulation of PBMC and synovial fluid mononuclear cells (SFMC ) then applied before or after stimulation of TNF-α ELISA kit to detect TNF-α the expression level changes . Results: 1. PBMC gene expression profiles compared 11 patients with SpA synovial tissue and 10 normal individuals , a total of four genes fluorescence intensity differences reached statistical significance , they are MMP- 3 , M -CSF , IL-7 and BAD proteins. 2.AS patients with peripheral serum samples and synovial fluid of patients with SpA can detect a certain concentration of MMP - 3 and M -CSF , and 15 patients with SpA were shut
