(Part I) In recent years, the study confirmed the vein or artery direct injection of bone marrow stromal cells can migrate to the brain damage zone, marrow stromal cell migration in vivo characteristics, but to migrate to the damaged region specific mechanism is unclear. The purpose of this study is to investigate the impact of the migration of chemokines and their receptors on bone marrow stromal cells. We first confirmed by immunofluorescence and RT-PCR method; followed by in vitro experiments confirmed bone marrow stromal cells in vitro expression chemokine receptor of CCR2, CX3CR1, CXCR4 and CCR5 chemokines MCP-1 and SDF-1 on bone marrow stromal cell chemotactic migration and its specificity. At the same time, we observed that the damage to the spinal cord injury District chemotactic factor MCP-1 and SDF-1 RNA levels compared with normal control has been improved, accompanied by changes in both visible and macrophages / microglia activation and by area of ??spinal cord injury intravenous injection of bone marrow stromal cells, considering the elevated MCP-1 and SDF-1 may be involved in chemotaxis in vivo expression of its specific receptor cells migrate to the damaged region, but can not rule out the involvement of other chemokines and their receptors. (Part II) In recent years, some scholars prove that transplanted bone marrow stromal cells promote axonal regeneration and improve motor dysfunction caused by spinal cord injury, but about how the transplantation of bone marrow stromal cells promote axonal regeneration, transplantation of cells and regeneration of axons The relationship is not yet clear. This study by immunofluorescence staining and immuno-electron microscopy methods, to discuss how to promote bone marrow stromal cell transplantation injury axonal regeneration. Two weeks when the bone marrow stromal cell transplantation, spinal cord injury area visible CFDA-SE labeled transplanted cells, the number of transplanted cells gradually decreased with time; eight weeks only see a small number of transplanted cells to survive the damage zone, consider a spinal cord injury in the region a large number of OX42 positive phagocytes / activated microglia and empty may affect the survival of the transplanted cells.
