The peony root active ingredient microemulsion percutaneous anti-inflammatory and analgesic effects and its potential basis

This research belonged to the project“The key technology of microemulsion vehicle system of the Traditional Chinese Medicine”established by State Administration of Traditional Chinese Medicine (the 15th key problems in science and technology---study of fundamental problems of appraisal of curative effect and safety of Chinese medicinal materials). The correlative contents have already applied for patents.1 Objectives1.1 Study the feasibility of microemulsion used as the vehicle system of the effective component of Chinese medicinal materialsPrepare the oral or percutaneous microemulsion on the basis of the characteristics of the microemulsion, observe the feasibility of microemulsion used as the vehicle system of the effective component, effective parts or prescription of Chinese medicinal materials. 1.2 Appraise the effectiveness of microemulsion used as the percutaneous vehicle system of the effective component of Chinese medicinal materialsPrepare the percutaneous microemulsion of TGP or TGP and AG (abbreviated as TA microemulsion), observe the percutaneous absorptive characters, and study their mechanism of action of anti-inflammatory and analgesic effect, as well as the mechanism of the promotion of the percutaneous absorptive characters by microemulsion.1.3 Study the correlative mechanism of pharmacological action of TGP, and the material basis of pharmacological action of white peony root.2 Methods2.1 Select the surface acting agent(SAA), the surface active assist-agent, the oil phase, the aqueous phase(hydrofacies) as well as their proportion through the phase diagram research and the skin absorption experiment of rats. Survey the morphous, particle diameter and distribution of the system by transmission electron microscope(TEM) and laser granularity meter. Survey the content of peoniflorin in microemulsion under the dissimilar condition by high performance liquid chromatography(HPLC) to monitor the stability.2.2 Collect the receiving fluid of percutaneous microemulsion by skin diffusion tester, determine the content of peoniflorin in collected fluid by HPLC, calculate percutaneous rate constant in vitro. Observe the change of skin dynamics of rats, investigate the absorption characteristic of skin. Compare the difference of percutaneous diffusion among the emulsion, microemulsion, micelle or watery solution, the influence of azone on the percutaneous result of peoniflorin.2.3 Gather the blood sample contained the medicine by carotid artery encheiresis of rabbits, survey the content of peoniflorin in blood by HPLC after using oral capsule or percutaneous microemulsion. Compare the change characteristics of the content of peoniflorin in blood between different preparation or different medications.
