Dystroglycan(DG) is an important struction for conection cell with cellular matrix. It composes two sub-unites:α-DG andβ-DG.α-DG is a peri-membrane reciptor combining with celluar matrix;β-DG is a transversing reciptor combining withα-DG. There is abnormal expression in some carcinoma such as breast and colon carcinoma in human being. The abnormal expression may have an important role on recurrent and metastisis in carcinoma. The porpuses of the project are going to study the abnormal expression ofβ-DG in oral squamous cell carcinoma, probe a new way of understanding recurrent and metastisis for oral squamous cell carcinoma.With immunohistochemistry, western blotting and RT-PCR, we probedβ-DG expression in normal skin, normal buccal mucosa, normal tongue mucosa, normal tongue musle, fresh sections of squomous cell carcinoma of the tongue and the buccal mucosa and cell lines of squomous cell carcinoma of the tongue and the buccal mucosa. We explored the changing ofβ-DG expression in cell lines when tissue inhibitors of metalloproteinases were added into the culture media. The result showed thatβ-DG expression was located in the base of normal suqmous cell epithelium.β-DG was expressed in muscle membrane of mucle fibre in normal tongue muscle and normal buccal muscle.β-DG expression dispeared in squmous cell carcinoma of the tongue and the buccal mucosa, BcaCD885 and Tca8113 cell lines. In Tca8113 cell lineβ-DG restored when tissue inhibitors of metalloproteinases were added into the culture media, in BcaCD885 the expression was not appears.β-DG was a single 43kDa band protein, on the contrary, there were 43kDa and 30kDa bands respectively in the squomous cell carcinoma and the cell lines. The 30kDaβ-DG disappeared in Tca8113 cell line when tissue inhibitors of metalloproteinases were added. The 30kDaβ-DG weakened in BcaCD885 cell line.According to the results, the dystroglycan complex in oral squamous cell carcinoma, which connect cell skeleton to extracellular matrix disrupted at the position ofβ-DG. The disrupted structure may induce the carcinoma cell invasion nearby normal tissue. And the disrupted structure may relate tumour invasion and metastasis in oral squmous cell carcinoma. Tissue inhibitors of metalloproteinases can stop or reduce the disruption.
