Preparation and Biological Activity of a Kind of Marine Sulfated Oligosaccharide

Sulfated polysaccharide is a kind of polysaccharide with sulfate groups, which is extracted from red algae. Studies have found that sulfated polysaccharides have relatively unique biological activity. In recent years, studies have shown that Sulfated oligosaccharides have some activities, such as anti-inflammation, antitumor, anti-oxidation, anti-virus and anti-coagulation, etc. The methods of preparation sulfated oligosaccharides are chemical degradation, physical degradation and enzymatic degradation.30Sulfated polysaccharidease-producing bacteria were isolated from various samples, including seawater, seaweed and soil (coming from sulfated polysaccharides producing station, Hainan province). By observing the clear circle size of strains and the stability of produced K-carrageenase, we chose the strain H3-2as the experimental strain finally. The thin-layer chromatography (TLC) results show that the degradation products by sulfated polysaccharidease from the strain H3-2comprise3main components. The K-carrageenase from H3-2was used to degradate sulfated polysaccharides. By the method of DNS and the thin-layer chromatography (TLC) testing, the optimum degradation reaction condition for sulfated polysaccharide was:0.5%sulfated polysaccharides completely dissolved in the heated distilled water as the degradation test substrate, the degradation reaction volume ratio of sulfated polysaccharides substrate and enzyme was1:0.5and the reaction time of degradation was48h. The low molecular weight coarse products of sulfated oligosaccharides were finally obtained, after the protein and salt was removed by Sevage and dialysis method respectively and freeze-drying.Using the blood from the heart of rabbit, determine the anticoagulant activity of sulfated oligosaccharides of laboratory homemade. Different concentrations of sulfated oligosaccharides can extend the time of CT, and has significant difference. APTT, PT and TT are independent indicators of the basic pathway of coagulation screening. Among them, the prolongation of APTT and TT has relationship to the inhibition of endogenous thrombin, while PT is the indicators of the extrinsic coagulation pathway, sulfated oligosaccharides can significantly prolong the clotting timeexperimental process in the APTT, PT and TT time, which has the most significant influence on APTT. And APTT was extension with the increases of sulfated oligosaccharides concentration. Thus it is speculated that the sulfated oligosaccharides may anticoagulant effect through the endogenous pathway.
