Population Structure and Genetic Diversity of the Tiger Frog (Hoplobatrachus Rugulosus) of Three Populations in East Area of China

Rugulosa (Hoplobatrachus rugulosus) belongs Amphibia Anura Ranidae Rana tigrina fork tongue species , widely distributed in southern China . Because of habitat loss and fragmentation and excessive human hunting , and its distribution range and number of wild populations are shrinking dramatically . In this study, the GenBank database and published seven pairs of the laboratory rugulosa specific microsatellites (Microsatellite) (?) | Things to study measured three tiger frog populations ( population in Fujian , Anhui and Jiangxi population population ) of genetic diversity and genetic structure . The results showed that: 1,7 microsatellite loci showed a higher genetic diversity. In 106 samples were detected 55 alleles at each locus allele number between 6-10 . The average alleles per locus was 7.86 . All loci were polymorphic information content is greater than 0.5 , the average PIC was 0.825 . Members point observed heterozygosity and expected heterozygosity at 0.637-0.840 and 0.811-0.872 , respectively , and the average observed heterozygosity and mean expected heterozygosity was 0.739 and 0.849 respectively . 2 , for all loci Hardy Weinberg equilibrium and linkage disequilibrium test showed that 61.9 % of the loci showed significant or extremely significant deviation Hardy Weinberg equilibrium (P lt; 0.01 or P lt; 0.05), and between points you did not show a clear linkage disequilibrium . P values ??were greater than 0.05 . 3, through the IAM, TPM or three mutants SMM model bottleneck effect of various groups tested , the results showed : Jiangxi, Anhui and Fujian three populations have experienced recent population bottleneck effect . 4 , using the fixation index (Fst) and Fisher's exact test were measured three degree of genetic differentiation among populations and found that microsatellite loci between groups did not occur in a variety of significant genetic differentiation , P values ??were greater than 0.001 . 5 , according to Nei's unbiased genetic distance and UPGMA dendrogram constructed show , Anhui and Jiangxi populations genetic distance , is 0.1146 , Anhui and Fujian population genetic distances , is 0.1437 . 6, three gene flow studies have shown : Nm values ??were greater than 1 , the gene flow between populations is higher. Meanwhile various geographic populations Fis more positive , indicating that individuals within each population there was widespread phenomenon of inbreeding .
