Studies on Ecotypical Male Sterile Line373S and Determination of Rapeseed Quality with NYDL-3000

Heterosis is one of the most important approaches to increase crop yield. Now, themain approaches for utilization of heterosis in rapeseed include cytoplasmic male sterility,genic male sterility, chemical hybridization agent and self-incompatibility. Our researchgroup found and developed an environment-sensitive male-sterile line373S from aspontaneous male sterile population. In order to conform the inherit model and thecytological characteristic, cytological observation and classical genetic investigation wereperformed for the373S utilize in the breeding programme. The results showed that：(1)373S was crossed with8C340,8C87,8C89respectively. The resulting F1were alsomale fertile. Among seven F2populations, one sterile plant was found in the one F2population, and one sterile plant was also occurred in F3population resulting from selfing ofan ecological male sterile plant in the F2population derived from the cross of8B123S-4and8C340-3, so we propose that373S was controlled by two pairs of gene, recessive nucleargene and thermo-sensitive gene, and the Ms had the dominant epistasis effect on the Ts.(2) Anther development of373S was normal before the tetrad stage,and becameabnormal at the beginning of tetrad stage. At early uni-nucleate stage, the degradation oftapetal cell became more obvious and the cytoplasm started to shrink, degrade. At thematurity stage, only the coat of sterile pollen grain was remained.(3) In order to overcome the import NIRS instrument’s weakness of the high pice,narrow practical range and optimizing difficulty of model, Near infrared Reflectancespectroscopy (NIRS) NYDL-3000designed by Oil Crops Research Institute of ChineseAcademy of Agricultural Sciences was compared with the traditional chemical methods inmeasuring three quality parameters of rapeseed in this study. The coefficient of the crossvalidation and RMSECV of oil content, total glucosinolates and erucic acid in rapeseedwere95.43﹪,98.53﹪,98.23﹪and0.0469,0.4198,0.1604between NIRS and chemical method, respectively. No obvious difference was found in the cross validation ofNIRS-3000and traditional chemical methods. External validation revealed the coefficientof correlation between NIRS and the chemical methods were0.8376and0.9517for totalglucosinolates and oil content and the compliance rate of erucic acid was99.12%betweenNIRS and the chemical method. There were no significant differences between the resultsobtained by NIRS and traditional chemical methods in the tests of cross validation andexternal validation. The result suggested that NIRS NYDL-3000analyzer can be applied todeterminate the quality parameters of oil content, total glucosinolates and erucic acid inearly breeding materials in rapeseed breeding programme.These results obtained has important reference value to the heterosis utilization anddetermination of the quality parameters of oil content, total glucosinolates and erucic acidin early breeding materials.
