Objective:To observe the effect of acupuncture on promoting the tear secretion of dry eye rabbits,and study on neurotransmitter and morphology of lacrimal gland,so as to explore the mechanism of acupuncture in increasing lacrimal fluid.Method:There are 42 selected Pure-bred New Zealand rabbits,6 of them were randomly selected as blank group, which did not be disposed.The other 36 were made into models using 1%atropine drops. Three days later the 36 rabbits got successful modeling, and they were divided randomly intotwo groups. The rabbits in the acupuncture group were given acupuncture with five points around the eye once a day with the needles retained for 15 minutes. They were treated for 25 days. No treatment was given in the model group. Various index of every group were measured in the vested time.Result:The schirmertest (SIT) in acupuncture group were significantly more than in the 3rd day of the experiment in any other time (P<0.05). there was significant statistical difference between the experiment 7 to 28 days and 1 day before (P<0.05).In the model group,1 day before the experiment and at each time point in the experiment are significantly different (P <0.05), it showed no statistical difference between the 3rd and lst,5th,7th,14th day (P>0.05).There was statistical difference between the two groups in the 5th,7th,14th day.Corneal fluorescein staining (FL), In acupuncture group, there are no statistical difference compared withl day before the experiment and 14th,28th day,The 3rd day in experiment and the other time points in addition to the 5th day were statistically significant (P<0.05).In model group,one day before the experiment and any time point after the experiment has significant difference (P<0.05). It showed no statistical difference between the 3rd and lst,5th,7th,14th day (P>0.05). There was statistical difference between the two groups in the 5th,7th,14th day.Acetylcholine (Ach) in the acupuncture group compared with the model in the 14th day, the differences are statistically significant (P=0.026<0.05), the other time points have no significant difference among the groups.Lactoferrin (LF) in each group, and between the two groups was no significant difference (P>0.05)It showed the expansion of columnar epithelial cells and abundant cytoplasm through the’ optical microscope in acupuncture group.Electronic microscope showed the outstretched rough endoplasmic reticulum in cytoplasm, Golgi and a large number of low-density Secretory vacuole after Acupuncture treatment. Optical and Electronic microscope showed marked morphological changes and active cell activities.Conclusion:Acupuncture can improve the concentration of neurotransmitter-Ach in the rabbit lacrimal gland, make the columnar epithelial cells of lacrimal gland life activities, promot tear secretion in dry eye rabbit.
