Objective:To investigate the effect of Sesamin on serum matrix metalloproteinase-13in atherosclerosis rabbits model. Methods:32Japanese white rabbits were devided into normal control group (n=8) and atherosclerotic group (n=24). Normal control group was fed with normal diet and atherosclerotic group was fed with high cholesterol diet. Atheroscleorsis was induced by injection of bovine serum albumin (250mg/kg)via ear-edge venous oncely. Atherosclerotic group was fed with high cholesterol diet for5weeks and devided into Saline group (n=8. fed with high cholesterol diet+Saline), Sesamin group (n=8. fed with high cholesterol diet+Sesamin). Atorvastatin group (n=8. fed with high cholesterol diet+Atorvastatin). At the day56. the concentrations of total cholesterol (TC) triglycerides (TG). high-density lipoprotein (HDL) and low-density lipoprotein (LDL) were evaluated with Oxidase method:the level of MMP-13was evaluated with Enzyme-linked immunosorbent assay (ELISA) method. The expression of MMP-13in aortic wall was evaluated with Immunohistochemistry staining. Results: The level of serum TC、TG、LDL in Saline group were higher than those in norml control group (P<0.01); The level of serum TC、TG、LDL in Sesamin group and atorvastatin group were significantly lower than those in Saline group (P<0.01); There were no significant differences of the level of TC、TG、LDL between Sesamin and atorvastatin group (P>0.05). The expression of MMP-13in aortic wall in saline group was significantly higher than normal control group (P<0.01); the expression of MMP-13in aortic wall in Sesamin group and atorvastatin group was lower than Saline group (P<0.05); the expression of MMP-13in aortic wall between Sesamin and atorvastatin group was no significant difference (P>0.05). the level of serum MMP-13in saline group was significantly higher than normal control group (P<0.01); the level of serum MMP-13in Sesamin group and atorvastatin group were significantly lower than saline group (P<0.01); the level of serum MMP-13in Sesamin group and atorvastatin group was no significant difference (P>0.05). Conclusions:(1) Sesamin should effectively reduce the level of serum TCn TG and LDL.(2) There was the expression of serum MMP-13in the atherosclerotic plaque.(3) Sesamin should suppress the oxidative repair of LDL and immunoinflammatery react through anti-oxidative effect, thus, reduce the secretion of MMP-13; Sesamin should reduce to expression of MMP-13in atherosclerotic plaque in aortic wall; Sesamin effectively inhibit the process of atherosclerosis.
