Objective: This purpose of this study was to investigate the effectsand possible mechanism of Polygonatum polysaccharide on human peripheral bloodlymphocyte apoptosis after a high-intensity exercise.Methods:14healthy men wererandomly divided into Polygonatum polysaccharide group(B) and Placebo group(A) andthen performed a high-intensity incremental treadmill exercise and recored exerciseduration,time and heart rate after respectively taking Polygonatum polysaccharideenteric-coated capsules with100mg/day and the same dose placebo for fourweeks.After cannulation of the cubital vein,blood samples of10mL were taken Beforeexercise,immediately after exercise.Apoptosis and expression of Bcl-2,Bax, Fas,FasLapoptosis-ralated proteins were measured by flow cytometry. Results:1.The aboveindices were no significant difference before exercise between the two groups.2.Ascompared with before exercise,the apoptosis increased significantly in two groupsimmediately after exercise,but B group was significantly lower than A group, thedifference between the two groups was significant.3.the expression of Bcl-2proteindepressed significantly in two groups, but B group was slightly lower than A group; theexpression of Bax protein increased significantly, Bcl-2/Bax depressed significantly intwo groups;no difference significant in the two groups.4. the expression of Fas proteinincreased significantly in two groups,no significant difference in FasL,but B group wasslightly lower than A group compared with before exercise; no difference significant inthe two groups.Conclusion:Polygonatum polysaccharide can antagonize a high-intensityexercise-induced apoptosis in human peripheral blood lymphocytes, makinglymphocyte aopotosis after exercise was significantly lower than the placebogroup,which was probably associated with increased Bcl-2protein,decreased Bax andFas, and increased Bcl-2/Bax,indicating that Polygonatum polysaccharide can protectlymphocytes through the mitochondrial pathway and death receptor pathway, so as toachieve the purpose of slow lymphocyte apoptosis.
