Objective:The study Gualou Xiebai Banxia Decoction myocard ial reperfusion injury induced apoptosis, and to explore it s possible mechanism.Methods:Left anterior descending coronary ligation constru ction ofmyocardial ischemia-reperfusion injury model,(1)Too bserve Gualou Xiebai Banxia Decoction improvement of reperf usion injury in rat heart rate disorders;(2) Observed in ra ts serum creatinekinase CK, creatine kinase isoenzyme CK-MBo f change;(3)The use of cardiomyocyteapoptosis by TUNEL assa y, immunohistochemical detection of myocardial Bcl-2, Baxan d P38MAPK protein expression changes;(4)Through an optical microscopeThe observation of myocardial ischemia andreperfu sion injury in rat cardiac morphological changes.Result:(1) The Gualou Xiebai Banxia Decoction can be effect ive against myocardial ischemia reperfusion injury caused b yabnormal heartrhythms, and compared with I/R group, thedif ference was statistically significant (P<0.01、P<0.05);(2) Gualou Xiebai Banxia Decoction can effectively inhibit myoca rdial cell injury caused by reperfusion, reduce CK、CK-MB lea kage,compared with I/R group, the difference was statistica lly significant (P<0.01、P<0.05);(3) Compared with model g roup,Gualou Xiebai Banxia Decoction can effectivelyinhibit t heoccurrence of myocardial reperfusin apoptosis,thereby redu cing reperfusion injury;(4) Gualou Xiebai Banxia Decoction group of Bc1-2protein expression wassignificantly increase d, a significant reduction of Bax protein expression, Compare d with I/R group, the difference was statistically significa nt (P<0.01、P<0.05);(5) The GualouXiebai Banxia Decoction c an improve effectively activity ofP38MAPK, thus inhibiting a poptosis;(6) By cell morphology, Gualou Xiebai Banxia Deco ction can effectively protect the cardiac tissue structure, inhibitionof reperfusion injury.Conclusion:(1) Gualou Xiebai Banxia Decoction with signifi cant improvements in reperfusion injury-induced arrhythmia;(2) Gualou XiebaiBanxia Decoction can effectively improve the organizational structure damage caused by myocardial re perfusion injury and reduce CK, CK-MB leakage protection of the myocardial tissue;(3) The effect mechanism of Gualou X iebai Banxia Decoction can effectively inhibit myocardial i schemia and reperfusion injury induced cardiac myocyteapopt osis may related to regulate Bcl-2/Bax protein expression a ndactivation of P38MAPK.
