Effect of annaopingchong tablets on aquaporin 9 and blood-brain barrier around hematoma after intracerebral hemorrhage in rats

Objective:To investigate the protection mechanism of Annaopingchong pill on intracerebral hemorrhage through the observation of the expression changes of peripheral tissue aquaporin9after intracerebral hemorrhage in rats and the effects on blood brain barrierMethods:To establish an intracerebral hemorrhage model in rats by injecting autologous arterial blood. The rats were randomly divided into sham-operation group,model group, mannitol group, Annaopingchong pill group,combined treatment group,All groups were administrated,Takeing the brain specimen, detect the expression of AQP-9by Using the immunohistochemical method inintracerebral hemorrhage after24h、48h、72h. To observe water content in brain tissue and blood brain barrier permeability changes, Using dry and wet weight method determination of water content in brain tissue and Evans blue (Evan’s blue,EB)content in the lateral intracerebral hemorrhage organization.Results:①The water content in brain tissue around the hematoma gradually increased in rats with intracerebral hemorrhage (P<0.05).Compared to the others in each time point, the water contents of the model group were increased, there were significant differences(P<0.01); Compared to the model gyoup in each time point, the water contents of Annaopingchong pill group, mannitol group and combined treatment were reduced accordingly,the difference was statistically significant.(P<0.05). Annaopingchong pill group combined mannitol group were best at72hours.②The expressions of AQP-9were increased in each time point in rats with intracerebral hemorrhage (P<0.01). The AQP9expressions of Annaopingchong pill group and combined treatment were reduced most remarkable。(P<0.05). Annaopingchong pill group combined mannitol group were best at48、72hours.③The permeability of blood brain barrier in24hours were increased obviously in lateral intracerebral hemorrhage organization,the color of the damage zone was darker blue than the normal,peaking at72hours.Group comparison: compared to sham-operation group, in brain tissue around the hematoma Evans blue (Evan’s blue,EB)content of model group in rats with intracerebral hemorrhage after24to72hours was significantly increased (P<0.05), peaking at72hours, the difference was statistically significant.Compared to model group,Evans blue (Evan’s blue,EB)content of Annaopingchong pill group was decreased in each time point (P<0.05)Conclusion:The water content in brain tissue around the hematoma,the AQP-9expressions and the permeability of blood brain barrier gradually increased in rats cerebral hemorrhage mode13days ago.Annaopingchong pill protects the blood-brain barrier to reduce brain edema probably by decreasing AQP-9expression levels.the effect which Annaopingchong pill combined Mannitol control the fastigium of brain edema with cerebral hemorrhage
