Effects of Compound Shuang-ling Antidiarrhea Power on Ulcerative Colitis and Its Effect on the HSP70Expression

Objective:To cure dextran sulfate sodium(DSS) induced ulcerativecolitis(UC) by injecting compound shuang-ling antidiarrhea power intostomach,examine the expression changes of HSP70in colon tissues andexplore the therapeutical effect and mechanisms of the drug on UC.Methods:A total of90clean SD rats were selected.10rats wererandomly chosen as normal control(NC),the rest80rats were used toconstruct the dextran sulfate sodiuminduced ulcerative colitis model.Changesin general and blood stool symptom were observed to test if the model wassuccessfully constructed.After developing this approach,80rats wererandomly divided into8groups of10rats,DT,DS,ST,SS,CT,CS,WT andWS,respectively.General circumstances including activity and eatingetc,Weight,stools and debris were observed,and the disease activity index(DAI) of all the rats was evaluated daily.The corresponding colon tissues ofthe rats were taken on day3and6,and all specimens were stained withHE.Then the pathological changes of colon tissues in rats were observedmicroscopically,and the tissue damage was evaluated.Meanwhile,theexpression level of HSP70was examined by Immunohistochemicalstain,Western-blotting,RT-PCR and so on.Results:1General circumstances of the ratsNormal control rats presented body weight gain, normal defecation,eating normal activities;DSS model group and saline group rats weightloss,the naked eye blood,eat less lazy;group CS and group WS in the rat beganto gain weight,stool without any blood,normal diet activities.According to thedisease activity index score,By three:the other group and normal control groupis all have statistically significant(P<0.05),model control group group compared with model physiological saline was not statistically significant(P>0.05),model control group respectively with compound shuang-lingantidiarrhea powder group and Wan-xin-mu-zai-kuai-kang group compared allhave statistically significant(P<0.05),model saline groups and compoundshuang-ling antidiarrhea powder group and Wan-xin-mu-zai-kuai-kang groupcompared all have statistically significant(P<0.05),compound shuang-lingantidiarrhea powder group and Wan-xin-mu-zai-kuai-kang group comparednot statistically significant(P>0.05);The sixth day between each group tocompare results with third days between each group compared with results onthe same.DT group and DS group there were no statistically significant group(P>0.05),ST group and SS group there were no statistically significant group(P>0.05),CT group and CS group compared are statistically significant(P<0.05),WT group and WS group compared statistical significance(P<0.05).2HE staining of rats bowel and the results of pathological damageThe colonic mucosa of normal control group rats show completestructure,simple columnar cells are arranged in order,and glands structurearrangement rules;The colonic mucosa of DSS model group and DSS salinerats show hyperemia,edema,or even missing,glandular hyperplasia,arrangingdisorderly,bowel columnar epithelium cell necrosis fall off,the cell boundariesnot clear,submucous infiltrate a large number of lymphocyte andneutrophils,show the typical inflammation change;The colonic mucosa of CTgroup and WT group rats show decreasing of edema congestion,submucosallymphocytes and neutropenia reduced;The damage structure of colonicmucosa of CS group and WS group rats have obvious recovered,the bowelcolumnar epithelial cells are arranged in order,and in only a few lymphocytesand neutrophils.According to the histologic damage assessmentstandards,statistical results show that,by three:the other group and normalcontrol group is all have statistically significant(P<0.05),model control groupgroup compared with model physiological saline was not statisticallysignificant(P>0.05),model control group respectively with compoundshuang-ling antidiarrhea powder group and Wan-xin-mu-zai-kuai-kang group compared all have statistically significant(P<0.05),model saline groups andcompound shuang-ling antidiarrhea powder group and Wan-xin-mu-zai-kuai-kang group compared all have statistically significant (P<0.05),compoundshuang-ling antidiarrhea powder group and Wan-xin-mu-zai-kuai-kang groupcompared not statistically significant(P>0.05);The sixth day between eachgroup to compare results with third days between each group compared withresults on the same.DT group and DS group there were no statisticallysignificant group(P>0.05),ST group and SS group there were no statisticallysignificant group(P>0.05),CT group and CS group compared are statisticallysignificant(P<0.05),WT group and WS group compared statisticalsignificance(P<0.05).3The expression of HSP70in the rat colon mucosaHSP70expression is the highest in normal control group rats colonicmucosa,nevertheless,in DSS model group and DSS model saline rats, theexpression of the HSP70bowel mucosa is significantly lower than the normalcontrol group;The expression of the HSP70in the CS group and WS groupcolonic mucosa is obvious enhancement compared to model group,but theexpression is still lower than the normal control group.Statistical results showthat, by three:by three:the other group and normal control group is all havestatistically significant(P<0.05),model control group compared with modelphysiological saline was not statistically significant(P>0.05),model controlgroup respectively with compound shuang-ling antidiarrhea powder group andWan-xin-mu-zai-kuai-kang group compared all have statistically significant(P<0.05),model saline groups and compound shuang-ling antidiarrhea powdergroup and Wan-xin-mu-zai-kuai-kang group compared all have statisticallysignificant(P<0.05),compound shuang-ling antidiarrhea powder group andWan-xin-mu-zai-kuai-kang group compared not statistically.significant(P>0.05);The sixth day between each group to compare results with third daysbetween each group compared with results on the same.DT group and DSgroup there were no statistically significant group(P>0.05),ST group and SSgroup there were no statistically significant group(P>0.05),CT group and CS group compared are statistically significant(P<0.05),WT group and WS groupcompared statistical significance(P<0.05).Conclusion:Compound shuang-ling antidiarrhea powder can cause thesymptoms of ulcerative colitis rat disappeared such as weight loss,eat less,having blood in one’s stool,can reduce the intestinal inflammatory cellsinvade,promote the rat mucosa of epithelial cells of appreciation andrecovery.The effects of Compound shuang-ling antidiarrhea powder mayassociated with the enhanced expression of HSP70,so that to play the role ofprotect epithelial cells of bowel mucosa and inhibit the inflammation.
