Application of multiplex polymerase chain reaction in the diagnosis of chronic infectious conjunctivitis

Objective:To explore the application value of multiplex PCR for the etiological diagnosis in chronic conjunctivitis.Methods:43cases of chronic infectious conjunctivitis (Group A,43eyes),42cases of dry eye(Group B,42eyes),41cases of typical autoimmune conjunctivitis (Group C,41eyes) and38cases of the normal control group (Group D,38eyes) were selected from July,2011to February,2012.All specimens was take from conjunctival sac. DNA was extracted by MPCR amplification (joined the bacterial and adenovirus primer), and the results of gel electrophoresis were observed with the gel imaging analysis system, and with bacterial culture, compared with the two results.Results:1. The positive rate of520bp and1004bp bands was27.9%and9.3%in Group A, respectively, there was significant differences between two results (X2=4.914, P=0.027), The positive rate of bacterial culture was25.6%, there was no statistically differences compared with the positive rate of520bp bands.(X2=0.059, P=0.808).2. The total positive rate of520bp and/or1004bp bands was32.6%,14.3%,12.2%and7.9%in from Group A-D, respectively.The total positive rate of group A was significant differences compared with the other groups (X2=3.943, P=0.047; X2=4.972,P=0.026; X2=7.400, P=0.007)Conclusions:MPCR assay can be rapidly and accurately identify the type of etiologic agent in chronic conjunctivitis, and it has a good application prospects in etiological diagnosis of chronic conjunctivitis.
