Effects of Epigallocatechin-3-gallate (EGCG) on Colorectal Cancer in Vitro

【Objectives】To study the inhibitory effect of epigallocatechin-3-gallate(EGCG) on colorectal cancer cells and the mechanism of inhibition of colorectal cancer.【Method】Colorectal cancer cells (LoVo cells、SW480cells、HT29cells、HCT-8cells) were cultured and the proliferation ability of cultured cells in varying concentration of EGCG (10ug/ml、20ug/ml、35ug/ml)were tested by MTT analysis,and the cell apoptosis and cells cycle were detected with flow cytometry,and the gene expressing of Notch1、Notch2、HES1and JAG1were detect with Real-time PCR.,the protein expressing of Notchl、Notch2、HES1and JAG1were detect with Western blot.【Results】the inhibition of EGCG(tested by MTT analysis) to colorectal cancer cells has dose dependence, and the inhibition of EGCG to HT29cancer cells has time dependence EGCG(detected with flow cytometry) can enhance the colorectal cancer cells’apoptosis rate. there is a Statistical significance compared the experimental group (35mg/L) with the control group (0mg/L).At the mean time, EGCG can block the cell cycle of SW480cells and LoVo cells at G0/G1phase, can block the cell cycle of HCT-8cells at G2/M phase and block the cell cycle of HT29cells at S phase,supress their proliferation.EGCG(detected by Real-time PCR) can up-regulate the gene expression of Notchl and down-regulate the gene expression of Notch2、 HSE1and JAG1.EGCG also can up-regulate the protein expression of Notchl and down-regulate the protein expression of Notch2.The down-regulation of the protein expression of HES1and JAG1were not significant.【Conclusion】EGCG can significantly inhibit the proliferation of the colorectal cancer cells. The mechanism of inhibition is changing cell cycle and inducing cell apoptosis.And the mechanism is related to EGCG can up-regulate the gene expression of Notchl and down-regulate the gene expression of Notch2、HSE1and JAG1.effect the gene expression of HES1and EGCG can up-regulate the protein expression of Notchl and down-regulate the protein expression of Notch2.
