[Objective]By using Western Blot, RT-PCR. and fluorescent quantitative PCR method to detect CIAPIN1’ expression in five kinds of colon cancer cell line Caco-2, COLO205, HT-29. HCT116. LOVO cells and with normal colonic epithelial cell NCM460contrast.exploring the role of the CIAPIN1gene in colon cancer development, and provide a theoretical basis for the further study of colon cancer targeted therapy and reversal of drug resistance[Methods]1. Training5kinds of colon cancer cells Caco-2, COLO205. HT-29, HCT116and LOVO, the normal colonic epithelium cell NCM460.2. Protein extraction of five kinds of colon cancer cell lines and normal colon epithelial cells, using the Western Blot detect CIAPIN1gene expression.3. Extraction of five kinds of colon cancer cell lines and normal colon epithelial cells mRNA by RT-PCR and fluorescenceQuantitative PCR analysis expression of CIAPIN1.4. By the three methods of detection, compared gene expression in colon cancer cell lines and normal colon epithelial cells of CIAPIN1.[Results]1. Western blot results show:The CIAPIN1gene was not expressed in normal colonic epithelial cells NCM460, showing relatively high expression in the five kinds of colon cancer cells.2. RT-PCR and quantitative PCR test results show:CIAPIN1gene expression in colon cancer cells than in normal colonic epithelial cells NCM460expression increased, that is. showing relatively high expression.[Conclusions]CIAPIN1in colon cancer cell lines on a relatively high, its expression may be involved in the development of colon cancer happen as an oncogene.
