Expression and Function of RASIP1in Hepatocellular Carcinoma

BACKGROUND:Hepatocellular carcinoma (HCC) is one of the five most common malignant tumor which has being severely threaten human health worldwide. High incidence of recurrence and metastasis is the key reason of poor prognosis of postoperative patients with HCC. There is no determinate method to resolve the problem of and metastasis of HCC. Therefore, investigating the molecular mechanism of recurrence and metastasis of hepatocellular carcinoma is benefit for the prognosis and postoperative therapy of patients. Many reasons, including decreased adhesion of tumor cells which may cause their falling off its primary place and spreading to other places, contribute to the recurrence and metastasis of HCC.Recently, there was research revealing that RASIPl can significantly increase the adhesion of the embryonic endothelial cell which suggest that RASIP1may correlate with the adhesion of the tumor cell.However, whether RASIP1play an important role in recurrence and metastasis in hepatocellular carcinoma is still unclear.OBJECTIVE:The expression levels of RASIP1mRNA and protein in HCC tissue and its cell lines were detected and analyzed in order to reveal the relationship between RASIPl and the tumorigenesis and advancement of hepatocellular carcinoma. METHODS:Exprssion level of RASIP1mRNA and protein in29hepatocellular carcinoma tissues and the corresponding adjacent noncancer liver tissues (ANLT), which got from the liver cancer laboratory of Xiangya hospital, Central South University and stored in-80℃,was detected by real-time PCR, RT-PCR and western blot,and that of HCC cell lines such as LO2、HEPG2. MHCC97-H and HCCLM3was detected using real-time PCR and western blot respectively and the expression trend in different cell lines was analysed.RESULTS:1.RASIP1expression was decreased significantly in HCC tissues compared with the corresponding ANLT;2. The expression level of RASIP1mRNA and protein in LO2was significantly higher than that of HCC cell lines (P<0.05); The expression level of RASIP1mRNA and protein in MHCC97-H and HCCLM3were significantly lower than that of HepG2(P<0O.05)CONCLUSION:1. HCC tissues have lower expression than that of ANLT;2. After analysed RASIP1level of HCC tissues and its cell lines, we speculate that RASIP1may suppress the recurrence and metastasis of HCC.
