The Prelinary Research of Caveolin-1and Signaltransduction and Apoptosis in Hepatocellular Carcinoma

Objective:To investigate the expression and significance of Caveolin-1ERK-1and Caspase-3protein in hepatocellular carcinoma(HCC),and to discuss its roles in oncogenesis of HCC, and to provide new approaches and experimental basis for improving the level of HCC diagnosis and prevention.Method:Thirty hepatocellular carcinoma specimens and corresponding adjacent tissues and ten normal liver tissues were collected at the surgical in Guangxi medical university tumor hospital.(both of them were confirmed by pathology) Before surgery they were not therapy through a radiotherapy or chemotherapy etc. The all samples were fixed by10%formaldehyde, embeded in paraffin and then made into4μm slice.The expression of Caveolin-1、ERK-1and Caspase-3protein by immunohistochemistry in10cases of normal liver tissue,and30cases of liver cancers and paracancer liver tissue.Result:(1)Immunohistochemistry showed that the expression rate of Caveolin-1protein in normal liver tissue, and paracancer liver and liver cancers tissue was90%,13.33%,10%respectively,while ERK-1and caspase-3were30%,56.67%,83.33%and90%,70%,40%respectively.And the difference had statistical significance(P<0.05).(2) The expression of ERK-1protein have correlation with clinical stage(P=0.019),while the expression of Caspase-3protein have correlation with tumor extrahepatic metastasis(P=0.031).However, the expression of Caveolin-1、ERK-1and Caspase-3protein have no clear correlation with age, size, number,AFP and recurrence etc age, portal cancerour thrombus, tumor recurrence,HbsAg etc.(3)The expression of Caveolin-1negatively correlated with ERK-1(r=-0.447, P=0.013),while the expression of Caspase-3had no correlated with Caveolin-1in hepatocellelar carcinoma (r=0.181, P=0.337).Conclusion:(1) Low or loss of Caveolin-1protein expression may be closely related with the occurrence and development of HCC, Caveolin-1mayhave inhibitory effect on the occurrence and development process of HCC.(2)Low expression of Caveolin-1protein may activate ERK-1protein,and lead to ERK signal transduction pathways lose normal coordinate the balance which is the important factor that could lead to HCC of the occurrence. The expression of ERK-1protein have correlation with clinical stage.(3)Low expression of Caspases-3could inhibit tumor cell apoptosis and promoting the tumor cell proliferation and cause liver cell carcinoma events. The expression of Caspase-3protein have correlation with tumor extrahepatic metastasis.(4)The expression of Caveolin-1negatively correlated with ERK-1,while the expression of Caspase-3had no correlated with Caveolin-1in hepatocellelar carcinoma.
