Effect of YISHENTANG on Cell Cycle Regulatory Protein p27 in the Kidney of Diabetic Rats

Objective: the use chain STZ (Streptozocin, STZ) intraperitoneal injection of a diabetic model induced SD rats; observe the effect of the Polyjuice Potion kidney soup \p27 expression, to angiotensin II receptor antagonists (Angiotensin receptor blocker, ARB) \Methods: Adult male SD rats to 41, were randomly divided into normal control group (CON) and diabetes modeling group. The diabetes model rats fasted but water 12 hours after intraperitoneal injection of 2% STZ-induced diabetic model, normal group were injected with equivalent doses of citrate buffer (0.1mol / l, pH4.3), and 72 hours after the measurement of blood glucose, such as blood glucose gt; 16.7mmol / L, and stable for 2-3 days operator modeling success. Successful modeling of the rats were then randomly divided into a the diabetic kidney soup treatment group (YS), diabetes valsartan group (ARB) and diabetic untreated group (DMN). The YS group was treated with concentrated drug solution equivalent crude drug 6.5g / kg body weight / day orally; the ARB group valsartan (trade name: Diovan), 20mg / kg body weight / day orally; CON and the the DMN group is given corresponding capacity of drinking water by gavage. Experiment began three weeks rats weekly measurement of body weight, five weeks after the measurement of body weight daily gavage dose based on body weight adjusted accordingly and continued treatment for 8 weeks; day before the end of the experiment, all rats were fasted for 12 hours but water measurements per rats fasting body weight, collected 24-hour urine record urine output measured 24-hour urine total protein, 24-hour urinary albumin; rats were killed with 10% chloral hydrate anesthesia, cardiac blood collection blood glucose, creatinine, blood urea nitrogen indicators. Take the left kidney, saline cleaning dry weighing, take the right kidney longitudinal incision half placed in 10% neutral buffered formalin fixed, paraffin-embedded, sliced ??Immunohistochemical detection of p27 expression, and using an image analysis system for quantitative analysis. Results: All diabetic rats were induced diabetes model group. Weight: the beginning of the experiment, each group had no significant difference; the end of the experiment, DM groups were significantly less than the CON group (P lt; 0.05), no statistically significant difference between DM group. Kidney relative weight: DM group were significantly greater than the CON group (P lt; 0.05), no statistically significant difference between DM group. Blood sugar: DM group were significantly higher than the CON group (P lt; 0.05), no statistically significant difference between DM group. Serum creatinine: no statistically significant differences among the groups. The BUN: DM group were significantly higher than in the CON group (P lt; 0.05) DM groups showed no significant difference. The 24h urine: diabetic group were significantly higher than the CON group (P lt; 0.05), DM groups showed no significant difference. 24h urine total protein content: CON group was significantly less than the DM group (P lt; 0.05), DM groups showed no significant difference. 24h urine albumin content: DM group were significantly higher than the CON group (P lt; 0.05), DMN group was significantly higher than the ARB group and YS group (P lt; 0.05), no statistically significant difference between the ARB group YS group . p27 expression: the the various DM group were significantly higher than the CON group (P lt; 0.05); DMN group was significantly higher than the ARB and YS group (P lt; 0.05); ARB the YS group, the difference was not statistically significant. Correlation analysis: p27 expression was a significant positive correlation with 24h urinary albumin (γ = 0.842, P lt; 0.05), and 24h urine total protein no statistical significance (γ = 0.162, P gt; 0.05). Conclusion: 1, kidney soup can reduce diabetic rat urinary albumin excretion, inhibit kidney p27 expression; the kidney soup prevention diabetic nephropathy and reduce kidney p27 expression. Kidney soup reduce urinary albumin content of diabetic rats, inhibition of renal expression of p27 role similar valsartan.
