Effects of Rhizoma Imperatae and Its Complex Soup on the Model of IgA Nephropathy in Rats

Objective: To establish a rat model of IgA nephropathy, explore of Rhizoma Imperatae its Compound soup on the rat model of IgA nephropathy rats and its mechanism. Methods: 50 male Sprague-Dawley rats, the random number method is divided into: normal control group, model group, Rhizoma Imperatae group, the Chine compound group, dexamethasone group (n = 10); addition to the normal control group, and the remaining 4 group with oral administration of bovine serum albumin (BSA) intravenous injection of staphylococcal enterotoxin B (SEB) and carbon tetrachloride (CCl 4 ) subcutaneous establish IgA nephropathy rat model, continuous administration four weeks, the normal control group and model group given normal saline orally;, respectively, in the experimental eight weeks, 10 weeks, 12 weekend to collect fresh urine of rats urine the erythrocyte (U RBC ) count, 24-hour urine protein (UP) determination; killed in the 12th week rat blood testing serum creatinine (Scr), blood urea nitrogen (BUN), using the enzyme-linked immunosorbent assay (Elisa) to detect the rats blood interleukin -2 (IL-2), platelet-activating factor (PAF) level; specimens from tissues of the rat kidney pathology (HE, PASM-Masson staining, immunofluorescence) check; using semi-quantitative immunohistochemical Western Blot assay content and protein expression levels of TGF-β1 in the rat kidney tissue. Results: The model group and normal control group comparison: rats in 8 weekend microscopic hematuria, microalbuminuria (P lt; 0.01), 10 weeks obvious gross hematuria, heavy proteinuria (P lt; 0.01) serum Scr, BUN levels significantly increased serum IL-2 levels were significantly reduced, the PAF TGF-β1 protein expression was significantly increased (P lt; 0.01); each treatment group compared with model group: U RBC , a 24-hour UP, Scr, BUN, PAF and TGF-β1 protein expression was significantly decreased serum IL-2 levels were significantly higher (P lt; 0.01); dexamethasone group, the Chine compound serum PAF Scr reduce the level of more than Rhizoma Imperatae group (P lt; 0.05); the Rhizoma Imperatae the group, the Chine Compound serum IL-2 levels than dexamethasone group (P lt; 0.05). Pathological changes: light microscope: normal rat glomerular morphology of the normal control group, model group, mesangial cells and mesangial matrix - severe hyperplasia, each treatment group compared with model group, mesangial cells significantly alleviated; immune fluorescence: normal control group the glomerular not found IgA fluorescence deposition, model group, IgA fluorescence was big lumpy deposition in the mesangial area IgA fluorescence of each treatment group and the degree of mesangial matrix strength are lower than the model group (P lt; 0.05). Conclusion: of Rhizoma Imperatae its Compound soup has a significant role in the treatment of IgA nephropathy rat model, and is superior to Chine Compound soup, hematuria, proteinuria can be significantly reduced, reducing the pathological changes and improve kidney function, the mechanism may be white Chine by stimulating the body to secrete IL-2, to reduce the generation of PAF, inhibition of the secretion and expression of renal TGF-β1 and play a role.
