Study on the Reaction of Peony Endogenous Phenolic Compounds and IAA

This study researched whether the reaction between endogenous phenoliccompounds of peony planets in vitro and IAA could produce new substances under the catalysis ofpolyphenol oxidase (PPO), and the influence of the reaction conditions such as the PH, thereaction time and so on. At the same time, take axillary bud of Feng Dan Bai which was one kindof peony as the experimental material, add different types or concentration of phenolic compoundsto the rooting medium of peony planets in vitro, and observe the condition of rootage. It alsostudied the influence of exogenous phenolic compounds on the enzymatic activity during theprocess of rooting, such as peroxidase (POD), polyphenol oxidase (PPO) and indole acetic acidoxidase (IAAO), and explored the influence of phenolic compounds on producing adventitiousroot, which could provide theoretical guidance for solving the difficult problems of producing rootof peony planets in vitro. The main results were as follows:1. Used the detected endogenous phenolic compounds of peony planets in vitro and IAA asraw material, researched the reaction between endogenous phenolic compounds of peony planetsin vitro and IAA under the catalysis of polyphenol oxidase (PPO), the results showed that bothcaffeic acid and catechol all reacted with IAA, and produced new substances. The HPLC chartshowed that it formed three new substances at5.572min and8.035min and9.045min after20hof caffeic acid and IAA’s reaction under the catalysis of PPO, and it produced three newsubstances at5.57min and8.040min and9.409min after20h of catechins and IAA’s reactionunder the catalysis of PPO, and it was found that both kinds of phenolics with IAA reacted mostcompletely when PH was5.0and the reaction reached a stable state after6h.2.Used Feng Dan Bai which was one kind of peony and easy to root as experimental material,it was to add up rooting situation after40days, for comparison of the rooting medium withoutphenolics, and added different types and concentration phenolic substances to the peony planets invitro rooting medium. Experimental results showed that the rooting rate and root index of test-tubeseedlings which were added the exogenous phenolic were higher than control, and the rooting rateand root index of test-tube seedlings which were added coffee acid of0.8mg.L-1to be processedwere highest.3.Used the peony planets in vitro treatment which were added the exogenous phenolic andhad the highest rooting rate and root index as experimental material, it was to research thedynamic change in the process of rooting of the activity of the POD, PPO, IAAO. With WPM and 3mg.L-1WPM+IAA for comparison, the results showed that the peony of test-tube seedlingshad the highest activity in the4d which was treated by caffeic acid and the activity wassignificantly higher than the one which wasn’t added the caffeic acid, and was on the rise after12d; PPO activity showed a trend of "up-down-up", the activity got the maximum in1d whenPPO was added into the medium, it was lower than1d in15d, and the activity was significantlyhigher than control; the change of IAAO activity was bigger, which got the maximum in3d and
