Objective:By revealing the specific microRNAs expression in the disease Chronic hepatitis B and its spleen-stomach damp-heat syndrome,we can explore the pathogenesis of chronic hepatitis B and find the molecular biological basis of the spleen-stomach damp-heat syndrome of chronic hepatitis B from the perspective of regulation of microRNAs.Methods:We collected10asymptomatic Chronic hepatitis B patients,10Chronic hepatitis B patients with spleen-stomach damp-heat syndrome and5healthy normal persons from the Chronic hepatitis B outpatient(2012.4-2012.12) in the Infectious Diseases Hospital of Chengdu and Sichuan Chinese medicine hospital referred to the "Viral hepatitis prevention and treatment programs(2000year)"、"Chronic hepatitis B prevention and treatment guidelines"(2005year) and "Viral Hepatitis TCM standards (Trial)"(1991year),took their peripheral blood10ml and separated the plasma to frozen pipes.marked and placed in a-80℃freezer for safekeeping.Took the samples to the Haibo Ho Biotechnology Co., Ltd in time for microarray experiment,and detected the microRNAs expression levels of each sample. Analysed the data in the SPSS19.0Software and the online SAS system,obtained the differentially expressed microRNAs profile in the disease Chronic hepatitis B and its spleen-stomach damp-heat syndrome.Choosed the significant differently (more than2-fold) expressed microRNAs to forecast their target genes with TargetScan (V5.2) software and describe their functions for further research.Results:(1)After the analysis,we found59microRNAs differently expressed in the patients with Chronic hepatitis B.in which30up-regulated and29down-regulated.Also there were32microRNAs significant differently (more than2-fold) expressed.among which12microRNAs up-regulated and with1107target genes,their functions involve nucleoside-triphosphatase regulator activity transcription factor activity、nucleotide binding、Protein binding and so on.Other20 microRNAs down-regulated and with1680target genes,their functions involve transcription factor activity、enzyme regulator activity、transferase activity、enzyme activator activity、gene silencing、tissue remodeling、generation of a signal involved in cell-cell signaling、nucleotide binding、Protein binding、Pigmentation and so on.(2)We found60microRNAs differently expressed in the Chronic hepatitis B patients with spleen-stomach damp-heat syndrome,in which26up-regulated and34down-regulated.Also there were23microRNAs significant differently (more than2-fold)expressed,among which10microRNAs up-regulated and with500target genes,their functions involve transcription factor activity、immune system development、transporter activity、cell adhesion and so on.Other13microRNAs down-regulated and with499target genes,their functions involve transcription factor activity、transferase activity. nucleic acid binding、Protein binding、transporter activity、immune system development and so on.Conclusion:There are specific microRNAs expression profiles in the disease Chronic hepatitis B and its spleen-stomach damp-heat syndrome,and talk about the new mechanisms of the occurrence of the disease Chronic hepatitis B and its spleen-stomach damp-heat syndrome,and build the foundation of depth study on spleen-stomach damp-heat syndrome of chronic hepatitis B based on the mode of "disease" and "TCM syndrome".
