The Influence of Bushenhuoxue Recipe on CD4~+CD25~+Regulatory T Cells in the Mice of Immunological Premature Ovarian Failure

Objective:Through observing the influence of Bushenhuoxue recipe on ovarian morphology, the ratio of CD4+CD25+Treg to CD4+and Foxp3mRNA of Immunological premature ovarian failure mice to discuss the molecular immunology mechanism of Bushenhuoxue method against autoimmune premature ovarian failure.Method:Using the mouse zona pellucida3peptide fragments to establish a model of autoimmune premature ovarian failure. The mice were i.g.administered with high-dose, low-dose of Bushenhuoxue Recipe comparing with Bujiale for four weeks, and then, optical microscope, micro-capillary cell analysis/counter, RT-PCR were used to assay the changes of ovarian morphology, the ratio of CD4+CD25+Treg to CD4+and the expression of Foxp3mRNA in the spleen of mice.The results:1.The indexes of ovarian weight in mice:Although there were no significant differences between the blank group and the other groups(P>0.05), the indexes of ovarian weight in the other groups were lower than the blank group,and the indexes of ovarian weight in the model group were the lowest.The indexes of ovarian weight in the Bujiale group were significantly higher than those in the model group (P<0.05). The indexes of ovarian weight in the low-dose group of Bushenhuoxue Recipe were higher than those in the model group,but there were no significant differences between the low-dose group of Bushenhuoxue Recipe and the model group, and it was identical between the high-dose group of Bushenhuoxue Recipe and the model group (P>0.05);there were significant differences between the Bujiale group and low-dose group of Bushenhuoxue Recipe (P<0.05),but it showed no differences between the Bujiale group and high-dose group of Bushenhuoxue Recipe; there were also no significant differences between the low-dose and high-dose group of Bushenhuoxue Recipe (P>0.05).2. The ovarian morphology of mice:The ovarian follicles were visible in the blank group, and there were no lymphocyte and leukocyte infiltration in the ovarian follicles and interstitial area. In the model group, the number of ovarian follicles was less than it in the blank group or there were no obvious follicles,meanwhile we could find lymphocyte and leukocyte infiltration in the ovarian follicles and interstitial area. Compared with the model group, the number of follicles in the low-dose group of Bushenhuoxue Recipe increased, and lymphocyte and leukocyte infiltration could be discovered in the follicles and interstitial area. More ovarian follicles were visible and a small number of lymphocyte and leukocyte infiltration can be found in the ovarian follicles and interstitial area in the Bujiale group and the high-dose group of Bushenhuoxue Recipe.3.The ratio of CD4+CD25+Treg to CD4+in the spleen of mice:There were no distinct differences between the blank group and the other groups(P>0.05); the ratio in the model group was higher than it in the other groups (P<0.05), what’s more, it was the lowest in the high-dose group of Bushenhuoxue Recipe; at last, there were no significant differences in the treatment groups(P>0.05).4. The expression of Foxp3mRNA in the spleen of mice:Compared with the blank group, the expression of Foxp3mRNA reduced in the model group and low-dose group of Bushenhuoxue Recipe(P<0.05), and it was the lowest in the model group,nonetheless there were no differences between the high-dose group of Bushenhuoxue Recipe and the blank group(P>0.05),and it was identical between the Bujiale group and the blank group; the expression of Foxp3mRNA in the Bujiale group was higher than it in the model group(P<0.05),in addition, there were no differences between the model group and the high-dose group of Bushenhuoxue Recipe (P>0.05),and it was identical between the model group and the low-dose group of Bushenhuoxue Recipe,as well as the treatment groups(P>0.05).Conclusion:1. The Bushenhuoxue Recipe may improve the function of ovarians by means of increasing the indexes of ovarians.2.The Bushenhuoxue Recipe may have the ability to modulate the immune function of autoimmune premature ovarian failure by means of regulating the level of Foxp3mRNA and the ratio of CD4+CD25+Treg to CD4+.
