Objective:To analyze how does huangqisijunzi decoction improve the tumor micro-environment by observing the anticancer effect, whole and local immune state of the tumor-burdened liver cancer mice. And to suggest the characteristics of treating with liver cancer by invigorating the spleen Qi, thus providing experimental basis of clinical reasonable application of cancer therapy.Method:We vaccinate1×106Hepal-6liver cancer cells on the orthotopic liver of C57BL/6mice to make the liver cancer mice. Observation:Firstly, to analyze how does different doses of decoction effect the tumor and immune system of the liver cancer model mice by observing the spleen, thymus index of suppressing rate of the models. Secondly, to analyze how does different doses of decoction effect the whole and tumor micro-environment immune state of the liver cancer model mice by observing the peripheral blood, spleen, tumor tissues and some normal tissues right beside the tumor tissues, as well as the proportion of their subsets CD4+,CD8+,NK1.1+cell and Treg cell.Result:1. Huangqisijunzi decoction could suppress the growth of the tumor of the tumor-burdened liver cancer mice, and the inhibition rate were all greater than30%, with statistical significance(P<0.05), and it can protect immune organs of the cancer mice with the significant difference of the decoction group compared to control group by obvious increased of thymus index (P<0.01)2.Huangqi sijunzi decoction on liver cancer a tumor-burdened mice peripheral blood, spleen,liver tissue beside the cancer CD3+CD4+T markup rate, CD3+CD8+T+markup rate, CD4+/CD8ratio were rising, with significant difference (P<0.05),which prompt huangqi sijunzi decoction can enhance tumor-burdened mice whole and liver cancer microenvironment of specificity anti-tumor immune function.3.Huangqi sijunzi decoction on liver cancer a tumor-burdened NK1.1markers in mice spleen, tissue adjacent to carcinoma rates are rising, with significant difference (P<0.05), which prompt huangqi sijunzi decoction can enhance tumor-burdened mice whole and liver cancer microenvironment of inherent antitumor immune surveillance immune function.4.Huangqi sijunzi decoction on liver cancer a tumor-burdened peripheral blood, spleen, liver tissue in mice, Treg cells in the liver tissue adjacent to carcinoma marker rate decreased, with significant difference (P<0.05),which prompt huangqi sijunzi decoction can reduce tumor-burdened mice whole and liver cancer microenvironment of immunosuppressive stateConclusion:huangqisijunzi decoction has inhibitory effect on liver cancer and can protect the immune organs,meanwhile,the decoction can promote immune function into full play. The whole immune and local tumor micro-environment of the tumor-burdened liver cancer mice behaved the same, And higher concentration will bring stronger immune function according to the mice
