Experimental Study of Shikimic Acid on Rats with Atherosclerosis

Objective:To observe the anti-atherosclerosis action of Shikimic acid, and the mechanism of its therapeutic effects. Methods:The acute toxicity effect of Shikimic acid in mice was detected and rat model with atherosclerosis was established by feeding fatty forage and vitamin D3(vitamin D3600,000IU/kg by celiac injection at one time) for12weeks.85healthy Wistar rats were selected and randomly divided into the following6groups:1.normal group (feed with normal diet);2.model group (feed with fatty-diet);3.Shikimic acid high dose group (feed with fatty-diet+Shikimic Acid:100mg/kg)4.Shikimic acid medium dose group (feed with fatty-diet+Shikimic Acid:50mg/kg).5.Shikimic acid low dose group (feed with fatty-diet+Shikimic Acid:25mg/kg).6. Simvastatin group (feed with fatty-diet+Simvastatin:7mg/kg).At the beginning of the experiment, the Shikimic acid groups or Simvastatin group was fed by gavage to the Shikimic acid or Simvastat for12weeks, while the normal group and model group were given the same volume of saline as Shikimic acid group. After12weeks, the indicator of hemorheology, blood lipid levels and antioxidant capacity were measured. And then the pathological changes of the aortic arch are observed by taking the sample of aorta.the serum lipid was detected.After the rats were killed, blood was collected to mearsure blood rheology and the levels of serum total eholesterol (TC), low density liPoProtein cholesterol (LDL-C), high density liPoProtein cholesterol (HDL-C), triglyeeride (TG), malonaldehyd (MDA), suPer oxide dismutase (SOD), and the pathomorphology checks in rats were carried. Results:(1) The LD50of Shikimic acid was more than7.2g/kg for mice in acute toxicity.(2) Shikimic acid significantly improves atherosclerotic mice hemorheological indicators, and it significantly reduces the blood viscosity (high, medium and low cut) of atherosclerotic mice, plasma viscosity, and hematocrit (P<0.05), compared to the model group.(3) The concentration of serum TG (P<0.05)、TC (P<0.01)、LDL-C (P<0.01) of atherosclerosis rats were significantly decreased, compared to the model group.(4) The activity of SOD increased highly;and the level of serum MDA was significantly decreased, compared to the model group (P<0.01).(5) Through Histological and morphological observation,the shikimic acid may protect the aortic endothelial cells (EC) of rats, and inhibit the hyperplasia of smooth muscle cells and the formation of elastic fiber. With Shikimic acid treatment, Pathomorphological changes in the aorta:the pathological changes lessened in the Shikimic acid group, compared to the model group, the aorta tissue obviously improved. Conclusion:The octagonal shikimic acid has the role of prevention and treatment of atherosclerosis, the main mechanisms of it are as follows:(1) Shikimic acid changes may suggest that the Shikimic acid can protect the aorta endothelial cell, improve the pathological change in aorta.(2) Shikimic acid may improve the hemorheological indicators in AS rats.(3) Shikimic acid can regulate the lipid metabolism, reduce serum TG, the TC, of LDL-c content, and improve the distribution of cholesterol in the body. The regulation of blood lipid disorders is one of the mechanisms of the shikimate anti As.(4) Shikimic acid may reduce the serum levels of MDA, increase the Serum level of SOD in AS rats, reduce oxidative damage, and inhibit of lipid peroxidation damage. The inhibition of lipid peroxidation may be the most important role of Shikimic acid anti As.(5) Shikimic acid changes may suggest that the shikimic acid can protect the aorta endothelial cell, inhibit the progress of AS, and improve the pathological change in aorta.
