The Inhibiting Effect of HaoBieYangYinRuanJian Prescription on the Synthesis of Extracellular Matrix in Rats with Alcoholic Liver Disease

ObjectiveTo study the effect of HaoBieYangYinRuanJian Prescription (HBYYRJP) on alcoholic liver disease in rats, and explore the possible mechanisms by inhibiting the synthesis of extracellular matrix of liver, to provide experimental basis of hepatic fibrosis for clinical use of HBYYRJP.Methods100healthy Wistar male rats, weighted200-240g, were taken. The model of alcohol liver disease in rats was established by feeding alcohol and fat diet, pyrozole, carbony iron. Rats were randomly divided into5groups:high dose of HBYYRJP (8.2g·kg’·d-1), low dose of HBYYRJF (2.59g·kg-1·d-1), colchicine group (0.09mg·kg-1·d-1), model group (alcohol:6.2g·kg-1·d-1), normal group (distilled water:1.2ml·100-1·g-1·d-1). The therapeutic groups:alcohol was given in the morning and drugs were given in the afternoon; the model group:alcohol was given in the morning and distilled water was given in the afternoon; the normal group:distilled water was given in the morning and in the afternoon. After10weeks, rats were put to death, and samples were taken. Pathological changes of hepatic tissue under light microscope were observed; death rate and weight were counted; hyp of hepatic tissue was measured by acid hydrolysis, and then converted to collagen protein; serum PCⅢ,Ⅳ-C, HA, LN were measured by radio-immunity; serum ALT, AST, TP, ALB were measured by spectrophotometer,GLOB=TP-ALB; serum MDA was measured by SPF; the levels/activity of MMP-2and MMP-9were measured by gelatin zymography; the level of hepatic tissue TGF-β1mRNA was measured by RT-PCR.Results1. Death rate and weight:The death rate in model group was significantly increased compared with normal group rats (P<0.01); the Death rate in therapeutic groups were significantly decreased compared with model group rats (P<0.01). The weight in model group was significantly decreased compared with normal group rats (P<0.01); the weight in therapeutic groups were significantly increased compared with model group rats (P<0.01). 2. Histological examination and pathological grading:Fatty liver and hepatitis were found in model group. An obvious improvement was found in hepatic tissue in HBYYRJF groups. The patho-grade in model group was significantly increased compared with normal group rats (P<0.01); the patho-grade in therapeutic groups were significantly decreased compared with model group rats (P<0.01).3. The level of collagen protein:The collagen protein in model group was significantly increased compared with normal group rats (P<0.01); the collagen protein in therapeutic groups were significantly decreased compared with model group rats (P<0.05).4. The levels of PCⅢ,Ⅳ-C, HA, LN:there was no significant difference of PC Ⅲ,Ⅳ-C, HA, LN in all groups (P>0.05).5. Hepatic functional parameter:The levels of ALT, AST, GLOB in model group were significantly increased compared with normal group rats (P<0.01); the levels of ALT, AST, GLOB in therapeutic groups were significantly decreased compared with model group rats (P<0.01). The levels of ALB, A/G in model group were significantly decreased compared with normal group rats (P<0.01); the levels of ALB, A/G in therapeutic groups were significantly increased compared with model group rats (P<0.01); there was no significant difference of TP in all groups (P>0.05).6. The level of MDA:MDA in model group was significantly increased compared with normal group rats (P<0.01); the MDA in therapeutic groups were significantly decreased compared with model group rats (P<0.01).7. The levels/activity of MMP-2, MMP-9:the brightness and area of hydrolytic strap of MMP-2, MMP-9in model group were significantly increased compared with normal group rats (P<0.01); the brightness and area of hydrolytic strap MMP-2, MMP-9in therapeutic groups were significantly decreased compared with model group rats (P<0.01).8. The level of TGF-β1mRNA:The level of TGF-β1mRNA in model group was significantly increased compared with normal group rats (P<0.05); the level of TGF-β1mRNA in therapeutic groups was significantly decreased compared with model group rats (P<0.05). Conclusions1. The model of alcoholic liver disease in rats was successfully established.2. HaoBieYangYinRuanJian Prescription can prevent alcohol-injured hepatic fibrosis in rats.3. The mechanisms may involve inhibiting the synthesis of extracellular matrix of liver by protecting liver cells, anti-inflamation, anti-lipid peroxidation, decreasing the levels/activity of MMP-2, MMP-9, inhibiting the expression of TGF-β1mRNA to resist hepatic fibrosis.
