Objective:To investigate the effect of thiazolidinediones on atherosclerosis (AS) of diabetic rats, and the expression of osteogrotegerin in atherosclerosis of diabetic rats. Methods Respectively to build a rat model of atherosclerosis for warfarin plus vitamin D3,a diabetes (DM) model for chain urine with cephalosporins.Methods:Take36male SD rats and randomly divided into6groups on average: AS group, DM group, AS+DM group, control group, AS+TZD group, AS+DM+TZD group. Measurements of metabolic parameters, von Kossa calcium dyeing, aortic calcium density and ALP (alkaline phosphatase) liveness, qRT-PCR for OPG (osteoprotegerin) mRNA.and immunohistochemistry for OPG were performed. ResultsResults:In AS group and AS+DM group,calcium deposition in aorta;aortic ALP liveness and calcium density of AS+DM group respectively rise3.70times,1.30times and3.64times,1.35times compared to AS group and DM group(P<0.05), OPG (osteoprotegerin) mRNA and expression decreased (P<0.05). Aortic ALP liveness and calcium density of AS+DM+TZD group were reduced18.03%and13.69%compared to AS+DM group(P<0.05), OPG (osteoprotegerin) mRNA and expression increased(P<0.05).Conclusion:Long-term thiazolidinediones treatment can attenuate the effect of diabetes on rat atherosclerosis, OPG (osteoprotegerin) mRNA and expression increased.OPG may be involved in the occurrence of inhibition atherosclerosis lesion.
