Experimenta Study on the Prevention Mechanism of Buzui Decoction on Acute Alcoholism Mice

Objectives:With the guidance of Chinese medicine theory, therapeutic effect and function mechanism of Buzui Decoction on acute alcoholism mice were studied, to offer a new thought and scientific basis for prevent acute alcohol poisoning by Chinese medicine drinks.Methods:SPF mice, male.20g±2g, were randomly divided into normal group, model group,(RU-21) group, and BZ Decoction (BZ) group. All groups were fasted but water for12hours before the experiment. Control group and experimental group preventive give the RU-21, BZ Decoction intervention respectively; normal group and model group fed normal saline by gavage. After30minutes, in addition to the control group was given normal saline. and the remaining to give alcohol of56°vol to establish a mice model of acute alcoholism. To observe alcohol tolerance time, plasma ethanol concentration, to examine the activity of ALT, AST and ALP in serum; the activity of TNF-a. IL-8. ADH, ALDH, P450, SOD and MDA in liver homogenate; to detect (3-EP, LENK in brain tissue homogenates, optical microscope observation of liver pathology change.Results:1. The alcohol tolerance time of BZ Decoction group (269.2±18.34min) is longer than model group and RU-21group,at the same time, the sober-up time of BZ Decoction group(284.40±27.0min) is shorter than the model group, the differences were statistically significant,P<0.01.2. The24h survival rate of BZ Decoction group mice compared with the model group significantly improved the survival rate of the two groups were94%,63%, and higher then RU-21group(77%). The differences of the live time were statistically significant,P<0.01.3. The content of AST. ALT and ALP in serum was lower in BZ Decoction group than those in the model group. The value is83.09±8.12(U/L).180.73±20.35(U/L)、 124.91±18.78(U/L), The differences of the live time were statistically significant, P <0.014. The content of TNF-α,IL-8,MDA in liver homogenate was lower in BZ Decoction group than those in the model group. The value is1.1164±0.05(ng/l)、0.2812±0.04(pg/ml)、3.75±0.4(nmol/ml).The differences of the live time were statistically significant, P<0.01.5. In BZ Decoction group, the liver damage caused by alcoholism reduced notability.6. BZ Decoction group in each time period (0.5h,1h,1.5h,2h,3h,6h).the plasma ethanol concentration (mg/ml) compared with model group, RU-21group is low, a concentration of28.16±1.09,31.12±0.95,32.74±0.97,37.47±1.04,30.02±0.86,26.98±1.17,P<0.01.7. BZ Decoction group of mice liver homogenates of ADH (U/ml), the ALDH (nmol/ml), cytochrome P450(nmol/ml), SOD (U/ml) activity compared with model group, anditsconcentrationwere0.295±0.037、93.73±15.37、215.42±14.99153.26±7.33, P<0.05.8. BZ Decoction group of mouse brain tissue homogenate, P-EP (U/ml) content LENK (nmol/ml) compared with the model group were1.541±0.46,1.682±0.03, P <0.05.Conclusion1. BZ Decoction has a more significant hangover wake-promoting effect,the possible mechanisms:①effectively reduce the alcoholism mouse model of the absorption of alcohol.②BZ Decoction can increase the ADH, the ALDH and P450. accelerate alcohol metabolism in the liver.③Its hepatoprotective mechanism and reduction of liver tissue homogenate of TNF-a, IL-8activity, reduce the inflammatory response.2. BZ Decoction has effect of hepatoprotective,the possible mechanisms:①by increasing the activity of SOD and decreasing MDA content, it reduces acute alcohol intoxication caused by liver oxidative damage.②reduction of liver tissue homogenate of TNF-a, IL-8activity, reduce the inflammatory response. 3. The original liquor of the BZ Decoction has a nice taste while showing significant hepatoprotective effects, and it is worth being popularized.
