"Yin Tianjing Fang" on multidrug resistance of human ovarian cancer cell line SKOV3/DDP in nude mice and the expression of LRP, GST- π effect

[Objective] Ovarian cancer is one of the common gynecologic malignancies, mortality is on the top of the female reproductive system cancer. The current treatment of ovarian cancer is still surgery, chemotherapy, radiotherapy, supplemented by surgery program is mainly cytoreductive surgery with platinum-based chemotherapy. Ovarian cancer cells develop resistance to chemotherapy drugs become an obstacle to the treatment of ovarian cancer, the study of Chinese and Western medicine ovarian cancer multidrug resistance mechanisms bcome more and more, the proportion of Chinese medicine in this study is gradually increased, and many studies have found that the active ingredients of traditional Chinese medicine can reverse tumor resistance, the tumor cells sensitive to chemotherapy, but few studies of traditional Chinese medicine to reverse drug resistance. After chemotherapy, clinical syndromes of patients are deficiency and qi deficiency, traditional Chinese medicine can effectively improve the symptoms of patients, but its mechanism is not clear. According to thinking of TCM, we bring forward hypothesis that " Ziyin tianjing fang " can regulate tumor resistance and verified by in vitro experiments, animal studies, to seek new ways to reverse tumor resistance.[Methods] Confirm the drug-resistance of SKOV3-DDP cells, and then establish the transplanted tumor models on nude mice, divide the mice into4teams including Platinum, Platinum and "ziyin tianjing fang","Ziyin tianjing fang". Calculating the inhibited rates, and then HE staining was used to examine the tissue structure, IH was used to detect the effect of "Ziyin tianjing fang" on the expressions of LRP. GST-Ⅱ.[Results]1."Ziyin tianjing fang and Platinum team" demonstrated significant inhibitory effects on the growth of tumor, the inhibitory rate was higher than the "Platinum team".2. Platinum team,"Ziyin tianjing fang and Platinum team","Ziyin tianjing fang" team could significantly down-regulate the expressions of LRP versus control, P<0.05. the expressions of "Ziyin tianjing fang and Platinum team"is lower than that of Platinum team,"Ziyin tianjing fang and Platinum team","Ziyin tianjing fang"team (P<0.05, P<0.05)."Ziyin tianjing fang"team is lower than Platinum team,(P<0.05).3. Platinum team,"Ziyin tianjing fang and Platinum team","Ziyin tianjing fang" team could significantly down-regulate the expressions of GST-π versus control, P<0.05, the expressions of "Ziyin tianjing fang and Platinum team" is lower than that of Platinum team,"Ziyin tianjing fang and Platinum team","Ziyin tianjing fang"team (P<0.05, P<0.05)."Ziyin tianjing fang"team is lower than Platinum team (P<0.05).[Conclusions]1. The prescription of "Ziyin tianjing" could enhance the inhibitory effects of Platinum on the growth of tumor.2."Ziyin tianjing fang" possibly reduced the drug-resistance through down-regulating the expressions of LRP and GST-π.
