BackgroundDrug addiction might involve various function changes in the central nervous system, more and more studies confirmed that the5-HT system is closely related to drug addiction,5-HT systemof the brain involved in motivation reward loop regulation maybecome5-HT neuralmechanisms of drug addiction.ObjectivesTo determinate the region changes in the level of5-HT and5-HIAA in nucleus accumbens before and after the establishment, withdrawal and reconstruction of the rat conditioned place aversion (CPA) model induced by morphine. To explore neurobiological mechanisms of rats addicted to morphine.Methods1. All69male SD rats were randomly divided into three groups, model group (MN group), and control group (MS group and SN group).2.Rats were injected with morphine,6.5days,10mg/kg, intraperitoneally (IP), once per day, naloxone injection,0.3mg/kg, IP, along with conditioned place aversion training, to develop the CPA model.3.The extracellular fluid of nucleus accumbens were collected using the method of microdialysis at timel (3-5days after microdialysis trocar implantation), time2(Day59:00AM,30min after morphine/saline injection), time3(Day5,90min after CPA behavior training), time4(Day6,40min after naloxone injection), time5(Day7,40min after CPA evaluation), time6(90min after last withdrawal behavior training), timc7(Day14,90min after reconstruction behavior training) respectively.4.5-HT and its metabolites in5-HIAA concentration in dialysate were measured by using high performance liquid chromatography with electrochemical detection.Results1. Chronic morphine injection and naloxone urged addiction collocation can be successfully established CPA rats model, experimental rats showed obvious aversion motive, the partner drug side showed obvious avoidance. CPA can success eliminated after seven days withdrawal training in rats, withdrawal rats can rebuild CPA again when exposed to CPA environment again.2. After chronic injection morphine, the level of5-HT and5-HIAA in nucleus accumbens of MN group and MS group were higher compare with that of control group(P<0.05); After the injection of naloxone, the concentration of5-HT and5-HIAA were significantly reduced(P<0.01); After withdrawal training, the concentration of5-HT and5-HIAA in MN group rise obviously(P<0.01). After CPA reconstruction, the level of5-HT and5-HIAA of MN group was obviously decreased(P<0.01).ConclusionThe5-HT level of rats nucleus accumbens may participate in morphine addiction withdrawal-induced aversion motivation. Metabolism and pass of5-HT to change the central motive is probably one of the clinical treatment of opioid addiction potential targets.
